The title compounds, CHO ( and ), are tetra-cyclic benzoates composed of a taxane ring with a fused dioxolane ring as the core skeleton. In compound , the five-membered dioxolane ring is essentially planar while the two cyclo-hexane rings and the cyclo-octane ring adopt chair and chair-chair forms, respectively, and there are three intra-molecular H⋯H short contacts. The corresponding ring conformations in are similar; however, one intra-molecular C-H⋯O inter-action and two H⋯H short contacts are observed, and the benzoyl and meth-oxy-methyl groups show orientational disorder.

Synthesis and Redox Chemistry of Co3E4 (E = P, As) Clusters.

Chemistry

January 2025

University of Regensburg, Inorganic Chemistry, Universitätsstrasse 31, D-93040, Regensburg, GERMANY.

The synthesis of the cluster complexes [(Cp'''Co)3(µ3,η2:η2:η2-E3)(µ3-E)] (E = P (3), As (4)) starting from the anionic triple-decker complexes [K(18cr6)(dme)2][(Cp'''Co)2(µ,η4:η4-E4)]- (E = P (1), As (2)) by electrophilic quenching with the Co dimer [(Cp'''CoCl)2] is reported. Both complexes show a distinct redox chemistry, which was first investigated by cyclic voltammetry. Subsequently, the monoanions [K(L)(sol)n][(Cp'''Co)3(µ3,η2:η2:η2-E3)(µ3-E)] (E = P, L = 18cr6, sol = dme, n = 2 (5), E = As, L = 2,2,2-crypt, n = 0 (6)), the monocations [(Cp'''Co)3(µ3,η2:η2:η2-E3)(µ3-E)][FAl] (E = P (7), As (8)) and the dications [(Cp'''Co)3(µ3,η3:η3:η3-E4)][TEF]2 (E = P (9), As (10)) could be realized experimentally and isolated in moderate to good yields.

De novo discovery of cyclic peptide inhibitors of IL-11 signaling.

Bioorg Med Chem

November 2024

Takeda Development Center Americas, Inc., 9625 Towne Centre Drive, San Diego, CA 92121, USA.

Interleukin-11 (IL-11), a member of the IL-6 cytokine family, has potential pro-inflammatory and pro-fibrotic roles in pulmonary, hepatic, cardiovascular, renal and intestinal disease pathogenesis, including oncogenesis. The potential for therapeutic intervention in these disease spaces has therefore made the IL-11 signaling axis an attractive target in drug discovery, and antibody inhibitors of IL-11 signaling are currently under evaluation in Phase I/II clinical trials. While lower molecular weight small molecule and peptide inhibitors may offer the potential for improved tissue penetration, developability and manufacturing cost compared with a protein therapeutic, reports of such chemical matter in the literature are limited.

Membrane-embedded CdaA is required for efficient synthesis of second messenger cyclic di-AMP.

Commun Biol

December 2024

Department of Biochemistry, Groningen Biomolecular Science and Biotechnology Institute, University of Groningen, Nijenborgh 3, Groningen, The Netherlands.

Cyclic di-adenylate monophosphate (cyclic di-AMP) is an important second messenger in microorganisms. Cyclic di-AMP regulates bacterial cell volume and turgor via control of potassium and compatible solute transport but is also involved in many other processes, including the activation of the metazoan innate immune response to bacterial infections. We compare the activity of full-length membrane-embedded CdaA, the enzyme that synthesizes cyclic di-AMP, with the water-soluble catalytic domain CdaA-DAC.

Nitrogen emissions up to the standard are a major challenge for wastewater treatment plants in alpine and high-altitude areas. The dosing of carriers can improve the nitrogen removal efficiency of the system at low temperatures; however, the mechanism of action of sludge and biofilm in nitrogen removal remains unclear. This study elucidated the internal mechanism of nitrogen removal via the function of microbial metabolism in sludge and biofilm at low temperatures.
