The Role of Exosomes in Cancer Progression and Therapy.

Biology (Basel)

January 2025

National Laboratory Astana, Center for Life Sciences, Nazarbayev University, Astana 010000, Kazakhstan.

Exosomes are small extracellular vesicles and are crucial in intercellular communication. Interestingly, tumor-derived exosomes carry oncogenic molecules, such as proteins and microRNAs, which can reprogram recipient cells, promote angiogenesis, and stimulate cancer pre-metastatic niche, supporting cancer growth and metastasis. On the other hand, their biocompatibility, stability, and ability to cross biological barriers make them attractive candidates for drug delivery.

Mesenchymal stem cell-mediated adipogenic transformation: a key driver of oral squamous cell carcinoma progression.

Stem Cell Res Ther

January 2025

Department of Stomatology, The Third Affiliated Hospital, Sun Yat-sen University, Guangzhou, 510630, China.

Background: Interaction between mesenchymal stem cells (MSCs) and oral squamous cell carcinoma (OSCC) cells plays a major role in OSCC progression. However, little is known about adipogenic differentiation alteration in OSCC-derived MSCs (OSCC-MSCs) and how these alterations affect OSCC growth.

Methods: MSCs were successfully isolated and cultured from normal gingival tissue, OSCC peritumoral tissue, and OSCC tissue.

Liquid biopsy is gaining importance in oncology in the age of precision medicine. Extracellular vesicles (EVs), among other tumor-derived indicators, are isolated and analysed from bodily fluids. EVs are secreted by both healthy and cancerous cells and are lipid bilayer-enclosed particles that are diverse in size and molecular makeup.

Tumor-derived extracellular vesicle (tEV)-associated RNAs hold promise as diagnostic biomarkers, but their clinical use is hindered by the rarity of tEVs among nontumor EVs. Here, we present EV-CLIP, a highly sensitive droplet-based digital method for profiling EV RNA. EV-CLIP utilizes the fusion of EVs with charged liposomes (CLIPs) in a microfluidic chip.

Development of Liver-Targeting αβ Exosomes as Anti-TGF-β Nanocarriers for the Treatment of the Pre-Metastatic Niche.

Biology (Basel)

December 2024

Departamento de Innovación Biomédica, Centro de Investigación Científica y de Educación Superior de Ensenada (CICESE), Ensenada 22860, BC, Mexico.

Liver metastases frequently occur in pancreatic and colorectal cancer. Their development is promoted by tumor-derived exosomes with the integrin αβ on their membrane. This integrin directs exosomes to the liver, where they promote a TGF-β-dependent pre-metastatic niche.
