Creating functional biological molecules de novo requires a detailed understanding of the intimate relationship between primary sequence, folding mechanism, and packing topology, and remains up to now a most challenging goal in protein design and mimicry. As a consequence, the use of well-defined robust macromolecules as scaffolds for the introduction of function by grafting surface residues has become a major objective in protein engineering and de novo design. In this article, the concept of scaffolds is demonstrated on some selected examples, illustrating that novel types of functional molecules can be generated. Reengineered proteins and, most notably, de novo designed peptide scaffolds exhibiting molecular function, are ideal tools for structure-function studies and as leads in drug design.

This case report describes metacarpophalangeal (MCP)- and metatarsophalangeal (MTP)-level amputation and reconstruction of all 4 paws of a young female domestic shorthair cat. All 4 paws, the ears, and the distal portion of the tail underwent severe frostbite injury resulting in tissue death. Following MCP and MTP amputation, weight-bearing surface reconstruction was achieved using metacarpal and metatarsal paw-pad advancement flaps.

Synovial exudation, creeping, and lubrication failure in natural cartilage under a long-term normal loading can be counteracted by a tribo-rehydration (sliding-induced rehydration) phenomenon. Hydrogels, as porous materials, can also restore interfacial lubrication and overcome creep through this strategy. At appropriate sliding velocities, water molecules at the interface contact inlet are driven by hydrodynamic pressures into the porous network to resist creep extrusion.

This clinical case report describes the reconstructive management of a child who developed a rare aggressive soft tissue infection - necrotizing fasciitis - complicating varicella skin lesions, with a synthetic dermal regenerative template - NovoSorb Biodegradable Temporizing Matrix - in conjunction with a split thickness meshed skin graft. The scarcity of this clinical case, the need to cover large skin defect promptly while facing infectious and pediatric challenges make it interesting to describe NovoSorb Biodegradable Temporizing Matrix in addition to split thickness meshed skin graft a novel treatment in necrotizing fasciitis of the child. A rapid surgical treatment followed by this reconstructive strategy achieved an acceptable functional and aesthetic result, with timely healing despite the severity and extensive surface area of the infection.
