Activity-dependent attenuations in extracellular spike amplitude have been shown to correlate with a decrease in the effectiveness with which somatic action potentials back-propagate into the dendritic arbor of hippocampal pyramidal cells. In this paper we demonstrate that activity-dependent attenuations in amplitude occur during behavior and that the amount of attenuation is reduced with an animal's experience in an environment. The observed reductions are caused by an animal's experience within a specific environmental context, are dependent on functional NMDA receptors, and are accompanied by an increase in the effective coupling of pyramidal cells and interneurons. These results provide an important step in linking together in vivo studies with in vitro data and suggest that mechanisms of plasticity engaged during behavior may be sufficient to alter the biophysical and integrative properties of hippocampal pyramidal cells.

Background: Closely related to the subfamily of proapoptotic proteins is the antiapoptotic protein bcl-2, which acts as an intracellular blocker of the mitochondrial apoptotic pathway. By inhibiting the action of effector caspases, as well as blocking the release of AIF and cytochrome C, Bcl-2 prevents regulated cell death and ensures survival in conditions of damage.
Method: The study was performed on Wistar rats, which were subjected to gravitational overloads (9g) in the caudocranial vector for 5 minutes twice a day for 28 days.

Background: Structural covariance analyses have identified macrostructural/morphological alterations to MRI-based networks in behavioral variant frontotemporal dementia (bvFTD), but microstructural/neuronal alterations to histology-based networks remain unexplored. We previously found greater neurodegeneration in layers and regions enriched for pyramidal neurons in bvFTD with tau (bvFTD-tau) compared to TDP-43 (bvFTD-TDP) pathology. Therefore, we hypothesized laminar networks of empirically connected pyramidal neurons are weaker in bvFTD-tau versus bvFTD-TDP.

Background: The initiation of amyloid plaque deposition signifies a crucial stage in Alzheimer's disease (AD) progression, which often coincides with the disruption of neural circuits and cognitive decline. While the role of excitatory-inhibitory balance is increasingly recognized in AD pathophysiology, targeted therapies to modulate this balance remain underexplored. This study investigates the effect of perampanel, a selective non-competitive AMPA receptor antagonist, in modulating neurophysiological changes in hAPP-J20 transgenic Alzheimer's mice.

Cortical neurons in brain slices display intrinsic spike frequency adaptation (I-SFA) to constant current inputs, while extracellular recordings show extrinsic SFA (E-SFA) during sustained visual stimulation. Inferring how I-SFA contributes to E-SFA during behavior is challenging due to the isolated nature of slice recordings. To address this, we recorded macaque lateral prefrontal cortex (LPFC) neurons in vivo during a visually guided saccade task and in vitro in brain slices.

Three-dimensional (3D) tissue culture models provide in vivo-like conditions for studying cell physiology. This study aimed to examine the efficiency of pyramidal microwell geometries in microfluidic devices on spheroid formation, cell growth, viability, and differentiation in mouse embryonic stem cells (mESCs). The static culture using the hanging drop (HD) method served as a control.
