Bacillus thuringiensis, isolate 114A, was used in toxicity experiments against the wild population ofthe olive pest Bactroceria oleae (Gmelin). In laboratory experiments, spores and crystals of the B.t. were delivered to the insects with the food. Longevity, oviposition period, number of eggs produced, and percent hatch were recorded. Olive fruits from the oviposition test were dipped into a suspension containing spores and crystals of B. thuringiensis 114A after the eggs were deposited. In field experiments, four to six sprayings per year of B.t. 114A isolate were applied for three successive years. It was found that, in addition to the longevity of B. oleae, the oviposition period, number of eggs and percent egg hatch decreased. Also, the percentage of pupation and emergence was reduced when olive fruits with eggs in their mesocarp were dipped in the solution of spores and crystals. Field applications with the toxins of 114A isolate of B. thuringiensis have resulted in significant protection of the olive production.

The nutrient germinant receptors (GRs) in spores of Bacillus species consist of a cluster of three proteins- designated A, B, and C subunits- that play a critical role in initiating the germination of dormant spores in response to specific nutrient molecules. Purpose: Flotation methods are widely used to detect oocysts/cysts of protozoans and eggs of helminths, except trematodes. However, details regarding the concentration and recovery rates of these parasites are poorly understood.

Methods: Using Eimeria tenella oocysts as a model parasite, the present study evaluated three check points: (1) the proportion of parasites that remain floating in flotation solution (sucrose or saturated saline) during centrifugation, (2) the proportion of oocysts that naturally float after addition of flotation solution after centrifugation, and (3) the rate of recovery on cover slips after completion of the flotation protocol.

Cry proteins, commonly found in Gram-positive soil bacteria, are used worldwide as aerial sprays or in transgenic plants for controlling crop pest populations and insect vectors. Via PCR analysis, a spore producing soil isolate (BV5) was speculated to encode a Cry gene. Partial nucleotide sequence of the amplified PCR fragment showed homology with the Cry8 genes present in GenBank.
