Interactions of Pyrene-Labeled Silica Particles.

Silica spheres were prepared by the Stöber method, and the method of labeling with pyrene, using aminopropylysilane as the linking agent, was optimized. Excimers between particles were formed when high concentrations of label were used. Both surfactant and polyelectrolyte addition were able to cause floc formation when added to suspensions of the silica, but only the surfactant changed the I(1)/I(3) ratio and diminished excimer formation. Synergetic Effects of Alanine and Glycine in Blob-Based Methods for Predicting Protein Folding Times.

The polypeptide PGlyAlaGlu was prepared with 20 mol % glycine (Gly), 36 mol % d,l-alanine (Ala), and 44 mol % d,l-glutamic acid (Glu) and labeled with the dye 1-pyrenemethylamine to yield a series of Py-PGlyAlaGlu samples. The fluorescence decays of the Py-PGlyAlaGlu samples were analyzed according to the fluorescence blob model (FBM) to obtain the number of amino acids (aa's) encompassed inside the subvolume of the polypeptide probed by an excited pyrene. An value of 29 (±2) was retrieved for Py-PGlyAlaGlu, which was much larger than for any of the copolypeptide PGlyGlu or PAlaGlu prepared with either Gly and Glu or Ala and Glu, respectively.

Characterization of the Interactions between an Unassociated Cationic Pyrene-Labeled Gemini Surfactant and Anionic Sodium Dodecyl Sulfate.

The gemini surfactant PyO-3-12, made of two dimethylammonium bromides joined by a propyl linker and bearing a dodecyl pendant on one side and a 1-pyrenemethoxyhexyl group on the other side, was employed to probe the interactions between positively charged PyO-3-12 and negatively charged sodium dodecyl sulfate (SDS). PyO-3-12 was selected for its ability to respond to the polarity of its local environment through the fluorescence intensity ratio / of the first-to-third fluorescence peaks of the pyrene monomer and the local pyrene concentration [] through the / ratio of the pyrene excimer-to-pyrene monomer fluorescence intensity. Furthermore, analysis of the fluorescence decays of aqueous solutions of PyO-3-12 and SDS yielded a measure of the internal dynamics, local concentration, and state (associated vs unassociated) of PyO-3-12 in solution.

The extensive use of organophosphorus pesticides in agriculture poses a high risk to human health and has boosted the demands for developing sensitive monitoring methods. Herein, we developed a facile and sensitive method for isocarbophos detection based on the remarkable fluorescence enhancement of pyrene during host-guest interaction of β-cyclodextrin polymer (β-CDP) and pyrene. The 3'-pyrene-labeled isocarbophos aptamer could be cleaved by exonuclease I to obtain free pyrene that was tagged on mononucleotides, which could enter the hydrophobic cavity of β-CDP, resulting in a prominent fluorescence enhancement.

The cationic gemini surfactant PyO-3-12 was designed to include two dimethyl ammonium groups, one dodecyl tail, and 1-pyrenemethyl hexyl ether tail into the structure of the surfactant. The pyrenyl label ensured that the fluorescence of pyrene could be employed to probe the behavior of PyO-3-12 at the molecular level. The introduction of the oxygen atom in the β-position to pyrene was found to be critical for restoring the sensitivity of the pyrenyl label to the polarity of its environment.
