We report the identification, genomic structure, chromosomal localization, and expression analysis of human frizzled-3 (FZD3), a 7-transmembrane receptor belonging to the frizzled family. The cDNA obtained from adult human brain shows 91% identity at the nucleotide level and 98% at the amino acid level to mouse frizzled-3 (fzd3). The FZD3 locus is located on chromosome 8p21, spans 48 Kb and its coding sequence is distributed in 6 exons intercalated by 5 introns. FZD3 is expressed in all analyzed human tissues, with quantitatively higher expression in the CNS and in urogenital structures.

Planar cell polarity (PCP) proteins localize asymmetrically to instruct cell polarity within the tissue plane, with defects leading to deformities of the limbs, neural tube and inner ear. Wnt proteins are evolutionarily conserved polarity cues, yet Wnt mutants display variable PCP defects; thus, how Wnts regulate PCP remains unresolved. Here, we have used the developing cochlea as a model system to show that secreted Wnts regulate PCP through polarizing a specific subset of PCP proteins.

Frizzled 3 receptor (FZD3) plays an important role in the homeostasis of the neural crest and its derivatives, which give rise to pigment-synthesizing cells, melanocytes. While the role for FZD3 in specification of the melanocytic lineage from neural crest is well established, its significance in the formation of melanoma, its associated malignancy, is less understood. In this study we identified FZD3 as a critical regulator of human melanoma tumorigenesis.

The orientation of mouse hair follicles is controlled by the planar cell polarity (PCP) pathway. Mutations in PCP genes result in two categories of hair mis-orientation phenotype: randomly oriented and vertically oriented to the skin surface. Here, we demonstrate that the randomly oriented hair phenotype observed in frizzled 6 () mutants results from a partial loss of the polarity, due to the functional redundancy of another closely related frizzled gene, Double knockout of and globally, or only in the skin, led to vertically oriented hair follicles and a total loss of anterior-posterior polarity.
