Two series of dendrimers containing a single ferrocene unit located in the focal point of these macromolecules have been synthesized and characterized. The first series of dendrimers has considerable lipophilic character, with tert-butyl ester groups located in their peripheral regions. In contrast, the second series of dendrimers was obtained by the hydrolysis of these peripheral ester groups, yielding water-soluble dendrimers with carboxylic acid groups in their surfaces. The electrochemical properties of these macromolecules were strongly affected by the dendritic groups attached to the ferrocene subunits. Host-guest interactions between the water-soluble dendrimers and the well-known receptor beta-cyclodextrin were also investigated. The dendritic groups were found to hamper markedly the formation of inclusion complexes between the cyclodextrin receptor and the dendrimer's ferrocene unit.

The conformation of a series of zero-generation polyamidoamine dendrimers end-labeled with four 1-pyrene-butyroyl, -hexanoyl, -octanoyl, -decanoyl, and -dodecanoyl derivatives, referred to as the PyCX-PAMAM-G0 samples with = 4, 6, 8, 10, and 12, respectively, was characterized in ,-dimethylformamide (DMF), dimethyl sulfoxide (DMSO), and aqueous solutions of 50 mM sodium dodecyl sulfate (SDS) or 50 mM dodecyltrimethylammonium bromide (DTAB). The conformation of the PyCX-PAMAM-G0 samples was determined from the global model-free analysis (MFA) of the fluorescence decays, which yielded the average rate constant (⟨⟩) for pyrene excimer formation (PEF) between an excited and a ground-state pyrenyl labels, with ⟨⟩ being proportional to the local concentration ([Py]) of the pyrenyl labels within the macromolecular volume; ⟨⟩-vs-[Py] plots yielded straight lines passing through the origin in DMF and DMSO, demonstrating that the internal segments of the dendrimers obeyed Gaussian statistics in these two solvents. In aqueous surfactant solutions, the hydrophobic pyrenyl labels induced the interactions of the PyCX-PAMAM-G0 dendrimers with the SDS and DTAB micelles.

Article Synopsis
  • * The study introduces new "fine-tuned" ChE inhibitors called PAMAM-calix-dendrimers, which were designed and synthesized with varying terminal fragments.
  • * Experimental results show that these dendrimers effectively inhibit human acetylcholinesterase and butyrylcholinesterase, with their activity linked to their structure, paving the way for future drug development.

This study introduces a series of water-soluble radical dendrimers (G0 to G5) as promising magnetic resonance imaging (MRI) contrast agents that could potentially address clinical safety concerns associated with current gadolinium-based contrast agents. By using a simplified synthetic approach based on a cyclotriphosphazene core and lysine-derived branching units, we successfully developed a G5 dendrimer containing up to 192 units of 2,2,6,6-Tetramethylpiperidinyloxy (TEMPO) radical. This synthesis offers advantages including ease of preparation, purification, and tunable water solubility through the incorporation of glutamic acid anion residues.

Childhood nephrotic syndrome (NS) is a serious disease affecting the health and quality of life of children, which is characterized by a series of pathophysiological changes due to the increased permeability of the glomerular membrane to plasma proteins. Low renal drug distribution and inefficient cellular uptake, resulting from cellular dysfunctions of filtration and internalization, are the main barriers to drug treatment in childhood NS, leading to deterioration in nephropathy. However, efficient therapeutic methods against childhood NS are still lacking in clinic.

Here, we present a modular synthesis as well as physicochemical and biological evaluation of a new series of amphiphilic dendrons carrying triphenylphosphonium groups at their periphery. Within the series, the size and mutual balance of lipophilic and hydrophilic domains are systematically varied, changing the dendron shape from cylindrical to conical. In physiological solution, the dendrons exhibit very low critical micelle concentrations (2.
