An endothelial nitric oxide synthase (eNOS) polymorphism in exon 7 (894 G/T) resulting in glutamate or aspartate, respectively, at position 298 on the protein is correlated with severity of cardiopulmonary diseases. Because glutamate and aspartate are considered to be conservative replacements, the polymorphism was thought to be a marker for a functional locus elsewhere in the gene. We now show in transfected cells, primary human endothelial cells, and human hearts, that eNOS with aspartate, but not glutamate, at position 298 is cleaved, resulting in the generation of 100-kDa and 35-kDa products. Recombinant or native eNOS was examined by immunoblotting either in lysates (COS7) or after partial purification over 2',5'-ADP-Sepharose and calmodulin-Sepharose. Immunoblotting after SDS/PAGE with a carboxyl-terminal antibody showed a single major protein band in the predicted position for eNOS at 135 kDa. An additional band at approximately 100 kDa was present only in the recombinant 298Asp eNOS and in the eNOS synthesized by primary cells and heart tissue with a G/T genotype. Using an eNOS amino-terminal-specific antibody, an immunoreactive band at approximately 35 kDa, corresponding to the residual N-terminal cleavage fragment, was observed in those cells with a T genotype. Thus, eNOS with aspartate but not glutamate at position 298 is cleaved, resulting in the generation of N-terminal 35-kDa and C-terminal 100-kDa fragments. Thus, the eNOS gene with polymorphisms at nucleotide 894 generates protein products with differing susceptibility to cleavage, suggesting that, in contrast to prior predictions, this polymorphism has a functional effect on the eNOS protein.

In athletes' competitions and daily training, in order to further strengthen the athletes' sports level, it is usually necessary to analyze the athletes' sports actions at a specific moment, in which it is especially important to quickly and accurately identify the categories and positions of the athletes, sports equipment, field boundaries and other targets in the sports scene. However, the existing detection methods failed to achieve better detection results, and the analysis found that the reasons for this phenomenon mainly lie in the loss of temporal information, multi-targeting, target overlap, and coupling of regression and classification tasks, which makes it more difficult for these network models to adapt to the detection task in this scenario. Based on this, we propose for the first time a supervised object detection method for scenarios in the field of motion management.

Structural and magnetic phase transitions in EuLaFe(BO) (x = 0, 0.18).

The crystal structures and hyperfine magnetic parameters of EuFe(BO) and mixed EuLaFe(BO) were studied over a wide temperature range in order to analyze correlations of the structural and magnetic features and the phase transitions in multiferroic compounds of the rare-earth iron borate family. The chemical compositions of the crystals are reported from X-ray fluorescence analysis. The crystal structures of EuFe(BO) and EuLaFe(BO) were determined using single-crystal X-ray diffraction in the temperature range 25-500 K.

Phytotoxicity Study of (Amino)imidazo[1,2-]pyridine Derivatives Toward the Control of , , and Weeds.

In this work, several imidazo[1,2-]pyridines were synthesized through the Groebke-Blackburn-Bienaymé three-component reaction (GBB-3CR), and their phytotoxicity was evaluated by the influence on the growth of wheat coleoptiles and three important agricultural seeds (, , and ) at test concentrations of 1000, 300, 100, 30, and 10 μM. A structure-activity relationship was established, showing the importance of halogen groups at the position of the attached aromatic ring and the presence of a cyclohexylamine group for greater activity. Post-modification of some GBB-3CR adducts was carried out, leading to imidazo[1,2-]pyridine-tetrazole hybrids, which were also evaluated in these bioassays.
