Simplified biomimetic peptide-based vehicle for enhanced tumor penetration and rapid enzyme-induced drug release.

Various nanodrug vehicles were well-designed with complicated functions for tumor therapy. However, the unsatisfactory tumor delivery efficiency and uncertain off-target release became the stumbling block of the nanodrugs on the way to the clinic. Inspired by efficient tumor targeting ability of albumin, we reported a simplified biomimetic peptide-based vehicle synthesized by copolymerizing L-glutamyl-L-lysine unit (EK dimer, an intrinsic surface peptide pair from albumin) with L-phenylalanine (F) to encapsulate doxorubicin (Dox).

Alzheimers Dement

December 2024

Johns Hopkins University, Baltimore, MD, USA.

Background: Blood exchange experiments in heterochronic parabionts or heterochronic plasma transfer have revealed that age-associated changes in blood composition contribute to neurogenesis and cognitive impairment in the elderly, and multiple blood-born pro-aging factors have been identified as causal factors to accelerate age-dependent brain changes. We previously demonstrate that PDGF-BB secreted by pre-osteoclasts in bone mediates cerebrovascular impairment during aging, but the bone-derived PDGF-BB in Alzheimer's disease (AD) progression is still uncertain.

Methods: MCI patients and two transgenic AD mouse models, App-ps1 and 5XFAD, were used to evaluated PDGF-BB level by ELISA measurement.

Alzheimers Dement

December 2024

Perelman School of Medicine, University of Pennsylvania, Philadelphia, PA, USA.

Background: Cardiometabolic disorders represent a pressing public health challenge and confer heightened susceptibility to age-related cognitive decline, including increased risk of developing Alzheimer's disease (AD). There is a strong genetic component underlying risks for these conditions, and human Genome-Wide Association Studies (GWAS) have associated the PPP1R3B genetic locus with both cardiometabolic dysfunction and AD. PPP1R3B protein is primarily expressed in liver hepatocytes, regulating liver glycogen metabolism.

Background: T cell mediated immunity is reported to play a pathogenic role in cardiac allograft vasculopathy (CAV) in heart transplant (HTx) patients. However, peripheral blood CD8 T cells have not been previously characterized in CAV. This study aimed to identify potentially pathogenic circulating CD8 T cell populations in high grade CAV patients using cellular indexing of transcriptomes and epitopes by sequencing (CITE-seq).

PINK1 modulates Prdx2 to reduce lipotoxicity-induced apoptosis and attenuate cardiac dysfunction in heart failure mice with a preserved ejection fraction.

Clin Transl Med

January 2025

Key Laboratory For Organ Failure Research, Ministry of Education of the People's Republic of China, Guangzhou, China.

Introduction: Heart failure with preserved ejection fraction (HFpEF) is a complex condition characterized by metabolic dysfunction and myocardial lipotoxicity. The roles of PTEN-induced kinase 1 (PINK1) and peroxiredoxin-2 (Prdx2) in HFpEF pathogenesis remain unclear.

Objective: This study aimed to investigate the interaction between PINK1 and Prdx2 to mitigate cardiac diastolic dysfunction in HFpEF.
