The significance of endogenous immune surveillance in acute lymphoblastic leukemia (ALL) remains controversial. Using clinical B-ALL samples and a novel mouse model, we show that neoantigen-specific CD4+ T cells are induced to adopt type-1 regulatory (Tr1) function in the leukemia microenvironment. Tr1s then inhibit cytotoxic CD8+ T cells, preventing effective leukemia clearance.

β-Lactams are the most widely used antibiotics for the treatment of bacterial infections because of their proven track record of safety and efficacy. However, susceptibility to β-lactam antibiotics is continually eroded by resistance mechanisms. Emerging multidrug-resistant (MDR) strains possessing altered alleles (encoding PBP2) pose a global health emergency as they threaten the utility of ceftriaxone, the last remaining outpatient antibiotic.

π-Expanded and N-Doped Fluoranthenes.

J Org Chem

January 2025

Universität Rostock, Institut für Chemie, A.-Einstein-Str. 3a, 18059 Rostock, Germany.

Novel π-expanded and N-doped fluoranthenes were synthesized and thoroughly investigated. These eight unsubstituted compounds are obtained in a multistep synthesis with CH activation as the last key reaction step. The structures vary in their position of π-expansion on the fluoranthene scaffold and the location of the pyridinic nitrogen atom.

SUMOylation involves covalent attachment of small ubiquitin-like modifier (SUMO) proteins to specific lysine residues on target proteins and regulates various aspects of their function. Sentrin-specific proteases (SENPs) are key players in both the conjugation reaction of SUMO proteins to their targets and the subsequent deconjugation of SUMO-conjugated substrates. Here, we provide the first comprehensive prenatal description of a lethal syndrome linked to a novel homozygous stop-gain variant in SENP7 c.

Traffic classification in SDN-based IoT network using two-level fused network with self-adaptive manta ray foraging.

Sci Rep

January 2025

Department of Computer Science, College of Computer and Information Sciences, Majmaah University, 11952, Al-Majmaah, Saudi Arabia.

The rapid expansion of IoT networks, combined with the flexibility of Software-Defined Networking (SDN), has significantly increased the complexity of traffic management, requiring accurate classification to ensure optimal quality of service (QoS). Existing traffic classification techniques often rely on manual feature selection, limiting adaptability and efficiency in dynamic environments. This paper presents a novel traffic classification framework for SDN-based IoT networks, introducing a Two-Level Fused Network integrated with a self-adaptive Manta Ray Foraging Optimization (SMRFO) algorithm.
