Peroxisome Proliferator Activator α Agonist Clofibrate Induces Pexophagy in Coconut Oil-Based High-Fat Diet-Fed Rats.

Biology (Basel)

December 2024

Laboratory of Veterinary Pathology, Cooperative Department of Veterinary Medicine, Tokyo University of Agriculture and Technology, 3-5-8 Saiwai-cho, Tokyo 183-8509, Japan.

Peroxisomes are crucial for fatty acid β-oxidation in steatosis, but the role of pexophagy-the selective autophagy of peroxisomes-remains unclear. This study investigated the effects of the peroxisome proliferator-activated receptor-α (PPARα) agonist clofibrate on pexophagy in a coconut oil-based high-fat diet (HFD)-induced hepatocarcinogenesis model. Rats were divided into four groups: control, clofibrate, HFD, and HFD with clofibrate.

Background: Previously, Japanese Environmental Mutagen and Genome Society/Mammalian Mutagenicity Study Group/Toxicogenomics Study Group (JEMS/MMS toxicogenomic study group) proposed 12 genotoxic marker genes (Aen, Bax, Btg2, Ccnf, Ccng1, Cdkn1a, Gdf15, Lrp1, Mbd1, Phlda3, Plk2, and Tubb4b) to discriminate genotoxic hepatocarcinogens (GTHCs) from non-genotoxic hepatocarcinogens (NGTHCs) and non-genotoxic non-hepatocarcinogens (NGTNHCs) in mouse and rat liver using qPCR and RNA-Seq and confirmed in public rat toxicogenomics data, Open TG-GATEs, by principal component analysis (PCA). On the other hand, the U.S.

Effects of maternal feeding of clofibrate on hepatic fatty acid metabolism in suckling piglet.

Background: Energy deficiency is a leading cause of the high pre-weaning mortality of neonatal piglets in the swine industry. Thus, optimal energy metabolism is of crucial importance for improving the survivability of neonatal piglets. The effective utilization of milk fat as primary energy is indispensably required.

Degradation mechanism and toxicity assessment of clofibric acid by Fe/PS process in saline pharmaceutical wastewater.

November 2024

School of Civil & Environmental Engineering and Geography Science, Ningbo University, Ningbo, People's Republic of China.

Article Synopsis
  • The study investigates the oxidation of clofibric acid (CA) using the Fe/persulfate (PS) process, focusing on how chlorine affects degradation and toxicity in saline pharmaceutical wastewater.
  • Results showed a high removal efficiency (70.91%) for CA at pH 3.0, but only low mineralization (20.99%), indicating that while CA can be removed, it doesn’t break down completely.
  • Moreover, the presence of chlorine led to the formation of toxic chlorinated by-products, raising ecological concerns around using the Fe/PS treatment in high-salinity environments.

The effect of aging on the repeated-dose liver micronucleus assay using N-nitrosodipropylamine, quinoline, and carbendazim.

The repeated dose liver micronucleus (RDLMN) assay has been sufficiently validated in terms of the numbers and types of chemicals studied. However, it remains unclear whether aging affects assay results. The OECD Test Guideline 407 (Repeated Dose 28-Day Oral Toxicity Study in Rodents) indicates that dosing should begin as soon as feasible after weaning and in any event before 9 weeks of age.
