The controversial concept of oral delivery of interferon (IFN) is reviewed in relation to possible mechanisms of action. Although there is a rational basis and some positive clinical results for believing that it may work, problems remain to understand which is the most effective IFN preparation and why. Owing to a minimal cost and lack of toxicity, the oral as well as other unconventional routes of IFN delivery deserve to be evaluated because they may become useful.

This study aimed to evaluate the dosimetric and clinical outcomes of flattening filter (FF) versus flattening filter-free (FFF) beams in head and neck cancer (HNC) patients treated with volumetric modulated arc therapy (VMAT). Twenty-four patients with 70/59.4/54 Gy dose prescribed in 33 fractions with simultaneous integrated boost treatment were retrospectively analyzed to compare treatment delivery efficiency, target coverage, sparing of organs at risk (OARs), and remaining volume at risk (RVR) in two HNC groups (nasopharyngeal and oropharyngeal).

Human papillomaviruses (HPVs) are responsible for the majority of sexually transmitted infections (STIs), some of which are oncogenic and can cause oropharyngeal or genital cancers. The HPV prevalence at the genital level varies according to the population studied but is higher in the seminal fluid of men suffering from idiopathic infertility than in the general population. The involvement of HPV in male infertility is supported by several studies suggesting that this virus can affect sperm quality by altering sperm DNA integrity, motility, number, viability, and morphology, and by inducing the production of anti-sperm antibodies (ASAs).

Background: We have previously developed a candidate therapeutic HPV DNA vaccine (pBI-11) encoding mycobacteria heat shock protein 70 linked to HPV16/18 E6/E7 proteins for the control of advanced HPV-associated oropharyngeal cancer (NCT05799144). While naked DNA vaccines are readily produced, stable, and well tolerated, their potency is limited by the delivery efficiency. Here we compared three different IM delivery strategies, including intramuscular (IM) injection, either with a needle alone or with electroporation at the injection site, and a needle-free injection system (NFIS), for their ability to elicit gene expression and to improve the potency of pBI-11 DNA vaccine.
