Specific cytochrome P450 enzymes show tissue-specific induction, and different regulatory units for expression of these enzymes have been identified. The regulation of the phenobarbital (PB)-inducible P450 genes has been relatively well characterized in terms of PB induction, but less so with regard to tissue-specific expression. CYP2B2 is not expressed in the rat lung, whereas cytochrome P450 2B1 (CYP2B1) is a dominating enzyme in the same tissue. The constitutive expression of CYP2B1 and CYP2B2 in liver is low, but inducible by PB, whereas the pulmonary expression of CYP2B1 is not induced by PB. This indicates utilization of different regulating mechanisms in the two organs. A gene construct consisting of the structural gene for LacZ coupled to a 1.3-kb 5' fragment of the rat CYP2B1 gene was used to generate transgenic mice in order to further elucidate the mechanism behind tissue-specific expression and PB induction of the CYP2B1 gene. Using reverse transcriptase-polymerase chain reaction on total RNA extracted from lung and liver tissue, a lung-specific transcription of the transgene was observed. Transcription of the construct was also observed in livers from PB-treated transgenic animals. By histochemical staining of lung sections with 5-bromo-4-chloro-3-indolyl-beta-D-galactopyranoside (X-gal), we demonstrated expression at the protein level in bronchiolar cells. In conclusion, our results revealed that the region extending to -1. 3 kb in the 5' flanking region of the CYP2B1 gene included sequences that could partly account for the lung-specific transcription of CYP2B1 and the hepatic induction of CYP2B1 transcription by PB.

Background: Arsenic emerges as most potent hazardous element ranked as number one in ATSDR (Agency for Toxic Substances and Disease Registry) list, can easily accumulate in fish, transported to humans via consumption and affect humans and aquatic organisms. Considering above, current experiment designed to evaluate cyto-genotoxicity and histological alterations induced by arsenic in Labeo rohita used as an animal model.
Methods: By applying complete randomized design sampling acclimatized individuals of Labeo rohita (10 batches of 10 each with triplicates) were exposed to nine definitive doses (12, 14, 16, 18, 20, 22, 24, 26 and 28 mgL) of arsenic in glass aquaria to determine 96-h lethal concentration (LC) of arsenic.

The epididymal fat is required for the maintenance of normal spermatogenesis, and the lipectomy of epididymal fat at different postnatal age results in disrupted expression patterns of several testicular steroidogenic enzymes. The current research examined the effect of epididymal fat lipectomy at different postnatal ages on expression of cytochrome 5α-reductase I, cytochrome P450 aromatase, androgen receptor (AR), and estrogen receptors (ER) α and β in the mouse testis after 2 weeks of the lipectomy. The lipectomy of epididymal fat at 2 months of postnatal age resulted in significant increases of expression levels of cytochrome 5α-reductase I, cytochrome P450 aromatase, AR, and ER α and β.

Fatty acid peroxygenases have emerged as promising biocatalysts for hydrocarbon biosynthesis due to their ability to perform C-C scission, producing olefins - key building blocks for sustainable materials and fuels. These enzymes operate through non-canonical and complex mechanisms that yield a bifurcated chemoselectivity between hydroxylation and decarboxylation. In this study, we elucidate structural features in P450 decarboxylases that enable the catalysis of unsaturated substrates, expanding the mechanistic pathways for decarboxylation reaction.
