'Genomics': buzzword or reality?

'Genomics' has become a widely used term, covering a range of approaches that make use of the newly acquired wealth of genome data (both on man and on a number of model organisms) to gain new insights and accelerate research. This review attempts to present a clear and balanced view of developments in this field, to describe the four major approaches that contribute to genomics (bioinformatics, genetic analysis of extended populations, large-scale expression studies, functional approaches), and to indicate applications in basic and pharmaceutical research.

"Big Data" has surpassed "systems biology" and "omics" as the hottest buzzword in the biological sciences, but is there any substance behind the hype? Certainly, we have learned about various aspects of cell and molecular biology from the many individual high-throughput data sets that have been published in the past 15-20 years. These data, although useful as individual data sets, can provide much more knowledge when interrogated with Big Data approaches, such as applying integrative methods that leverage the heterogeneous data compendia in their entirety. Here we discuss the benefits and challenges of such Big Data approaches in biology and how cell and molecular biologists can best take advantage of them.

"Endocrine disruption" is a public and political buzzword that has and is still receiving high media attention. Based on the latter, numerous tiered testing strategies have evolved that should ensure that humans will not run a health risk due to the voluntary or involuntary exposure to endocrine active compounds (EAS). An analysis of the currently available knowledge on EAS mediated endocrine disruption in humans demonstrates that there are very few EAS that causally induce endocrine disruptive effects.

To what degree has targeted therapy succeeded in acute myeloid leukemia (AML)? Targeted therapy has become a buzzword, with its meaning lost from overuse. In chronic myeloid leukemia (CML), gastrointestinal stromal cell tumor, and a small subset of patients with non-small cell lung cancer, a validated target has been identified and a highly specific therapeutic agent has been developed. Targeted therapy generally requires a pathophysiological Achilles heel in a tumor that can be exploited by nontoxic therapy.
