Parkinson's disease (PD) is a complicated neurodegenerative disease that is the most prevalent severe movement disorder worldwide. The research includes studying the levels of hydrogen sulphide (H2S) and cystathionine γ-lyase (CSE) with some biochemical parameters in the serum of patients with PD in Mosul City (Iraq), which include Serotonin (SERT), dopamine (DA), sphingomyelin (SM), vitamin B12, Acetylcholine esterase (AChE), monoamine oxidase (MAO), creatine kinase (CK), aspartate aminotransferase (AST), alanine aminotransferase (ALT). Samples reached (100), which included: (40) for the Parkinson's patients group, and (60) for the control group.

Iontronic sensors based on confined space have garnered significant attention due to their promising applications, ranging from single-cell analysis to studies. However, their limited sensitivity has constrained their effectiveness in studying molecular information during physiological and pathological processes. Here, we demonstrate an electrolyte-gated ionic transistor (EGIT) by integrating the confined ion transport behavior in a double-barreled micropipet with an electrolyte-gated transistor configuration, achieving highly sensitive and selective sensing.

Drug inhibition and substrate transport mechanisms of human VMAT2.

Nat Commun

Vesicular monoamine transporter 2 (VMAT2) is crucial for packaging monoamine neurotransmitters into synaptic vesicles, with their dysregulation linked to schizophrenia, mood disorders, and Parkinson's disease. Tetrabenazine (TBZ) and valbenazine (VBZ), both FDA-approved VMAT2 inhibitors, are employed to treat chorea and tardive dyskinesia (TD). Our study presents the structures of VMAT2 bound to substrates serotonin (5-HT) and dopamine (DA), as well as the inhibitors TBZ and VBZ.

In recent years, modulation of microglial phenotype transformation has emerged as a promising strategy for treating central nervous system disorders. Aurantii Fructus Immaturus (Zhishi), a traditional Chinese medicine with versatile applications, contains p-Synephrine (p-SYN) as its principal bioactive compound, recognized for its anti-inflammatory efficacy. However, the molecular mechanisms underlying these effects remain unclear.

Proximity-dependent biotinylation coupled with mass spectrometry enables the characterization of subcellular proteomes. This technique has significantly advanced neuroscience by revealing sub-synaptic protein networks, such as the synaptic cleft and post-synaptic density. Profiling proteins at this detailed level is essential for understanding the molecular mechanisms of neuronal connectivity and transmission.
