Cell-Free Gene Expression from DNA Brushes.

Linear double-stranded DNA polymers coding for synthetic genes immobilized on a surface form a brush as a center for cell-free gene expression, with DNA density 10-10 fold higher than in bulk solution reactions. A brush localizes the transcription-translation machinery in cell extracts or in cell-free reconstituted reactions from purified components, creating a concentrated source of RNA and proteins. Newly synthesized molecules can form circuits regulating gene expression in the same brush or adjacent ones.

Autism spectrum disorders (ASD) have long-term implications on functioning at multiple levels. In this perspective, we offer a brainstem-informed autism framework (BIAF) that traces the protracted neurobehavioral manifestations of ASD to early life brainstem dysfunctions. Early life brainstem-mediated markers involving functions of autonomic/arousal regulation, sleep-wake homeostasis, and sensorimotor integration are delineated.

Three-dimensional tuning of idiophone bar modes via finite element analysis.

The timbre of marimba and other idiophone bars can sometimes be polluted by untuned torsional modes, leading to substandard instruments or rejected materials. Makers have complained of problems with these untuned modes over a specific range of notes. Marimba, vibraphone, and similar idiophone bars are tuned by carving one side of the bar to bring up to three flexural modes into harmonic relationships.

Cohesive floodplain sediment and vegetation are both thought to cause meandering river patterns. Our aims are to compare the isolated and combined effects of mud and vegetation on river planform and morphodynamics in the setting of intermediate-sized valley rivers. We use a numerical model for century-scale simulation of flow, sediment transport and morphology coupled with riparian vegetation settlement, growth and mortality as functions of species traits on which flow resistance depends.
