How close is autophagy-targeting therapy for Alzheimer's disease to clinical use? A summary of autophagy modulators in clinical studies.

Alzheimer's disease (AD) is a neurodegenerative disorder clinically characterized by progressive decline of memory and cognitive functions, and it is the leading cause of dementia accounting for 60%-80% of dementia patients. A pathological hallmark of AD is the accumulation of aberrant protein/peptide aggregates such as extracellular amyloid plaques containing amyloid-beta peptides and intracellular neurofibrillary tangles composed of hyperphosphorylated tau. These aggregates result from the failure of the proteostasis network, which encompasses protein synthesis, folding, and degradation processes.

Background: With increasing evidence supporting three-dimensional (3D) automated breast (AB) ultrasound (US) for supplemental screening of breast cancer in increased-risk populations, including those with dense breasts and in limited-resource settings, there is an interest in developing more robust, cost-effective, and high-resolution 3DUS imaging techniques. Compared with specialized ABUS systems, our previously developed point-of-care 3D ABUS system addresses these needs and is compatible with any conventional US transducer, which offers a cost-effective solution and improved availability in clinical practice. While conventional US transducers have high in-plane resolution (axial and lateral), their out-of-plane resolution is constrained by the poor intrinsic elevational US resolution.

Perfluorooctane sulfonate (PFOS) exerts adverse effects on neuronal development in young population. Limited evidences have shown that early-life PFOS exposure holds a potential risk for developing age-related neurodegenerative diseases such as Alzheimer's disease later in life. The present study investigated the effects of lactational PFOS exposure on cognitive function using one-year-old mice.

Distinct subcellular localization of tau and alpha-synuclein in lewy body disease.

Acta Neuropathol Commun

January 2025

Center for Neurodegeneration and Experimental Therapeutics, Department of Neurology, University of Alabama at Birmingham, Birmingham, AL, USA.

Lewy bodies and neurofibrillary tangles, composed of α-synuclein (α-syn) and tau, respectively, often are found together in the same brain and correlate with worsening cognition. Human postmortem studies show colocalization of α-syn and tau occurs in Lewy bodies, but with limited effort to quantify colocalization. In this study, postmortem middle temporal gyrus tissue from decedents (n = 9) without temporal lobe disease (control) or with Lewy body disease (LBD) was immunofluorescently labeled with antibodies to phosphorylated α-syn (p-α-syn), tau phosphorylated at Ser202/Thr205 (p-tau), or exposure of tau's phosphatase-activating domain (PAD-tau) as a marker of early tau aggregates.

Rapid Recognition and Monitoring of Multiple Core Biomarkers with Point-of-Care Importance through Combinatorial DNA Logic Operation.

The early diagnosis of a disease relies on the reliable identification and quantitation of multiple core biomarkers in real-time point-of-care (POC) testing. To date, most of the multiplex photoelectrochemical (PEC) assays are inaccessible to home healthcare due to cumbersome steps, long testing time, and limited detection efficiency. The rapid and fast-response generation of independent photocurrent for multiple targets is still a great challenge.
