Purpose: The expression of MAGE-A9 in vocal fold leukoplakia (VFL) tissues and the mechanism underlying its role in the malignant transformation of VFL remain unclear. This study investigated the role of MAGE-A9 in the proliferation, migration, and invasion of VFL epithelial cells as well as the underlying regulatory mechanism.

Methods: MAGE-A9 expression in VFL tissues was detected by immunohistochemistry.

The importance of vocal cord leukoplakia (VCL) in the etiology and progression of laryngeal carcinoma has gained increasing recognition. However, research into the mechanisms of laryngeal precancerous lesions such as VCL, has been hampered by the small size of VCL epithelial cells and their limited culture lifespan. In this study, we enhanced the primary culture protocol for VCL epithelial cells and introduced simian virus 40 Large T to establish an immortalized cell line, designated hVCL-MSDEP01.

Article Synopsis
  • - This study evaluated the expression of glucose transporter (GLUT)-1 and hexokinase (HK)-II in vocal cord leukoplakia (VCL) and explored its clinical importance using samples from various throat conditions over a four-year period.
  • - Results showed significant differences in factors like sex and smoking between VCL and vocal cord polyp patients, with GLUT-1 and HK-II expressions being higher in laryngeal carcinoma cases, particularly in severe dysplasia.
  • - Both GLUT-1 and HK-II levels increased with the severity of dysplasia, indicating that their expression could serve as biomarkers for assessing the progression of VCL and its potential transformation to cancer.

Article Synopsis
  • The study aimed to investigate the impact of acidic pepsin on Glut-1 expression and glycolytic activity in laryngeal cancer cells, focusing on its role in cell growth and migration.
  • Researchers treated laryngeal carcinoma cells with acidic pepsin and evaluated various metrics including Glut-1 levels, glucose uptake, and cell viability using multiple assays.
  • Results revealed that acidic pepsin significantly increased Glut-1 expression and glycolytic activity, promoting cancer cell growth and migration, effects that could be reversed with a glycolytic inhibitor and Glut-1 siRNA.

Article Synopsis
  • Upregulation of PD-1/PD-L1 lets cancer cells evade the immune system by disrupting T-cell function, making it challenging for immunotherapy to be effective in many patients with advanced cancer.
  • Radiotherapy can enhance the antitumor effects by promoting T-cell recruitment and increasing PD-L1 expression, while also altering tumor metabolism, which is linked to cancer immune evasion.
  • PET/CT imaging offers a non-invasive way to monitor PD-1/PD-L1 levels in tumors, providing advantages in tracking these markers over traditional immunohistochemistry methods to better inform treatment strategies.

Article Synopsis
  • The study aimed to explore how H/K ATPase affects the growth of pepsin-induced vocal cord leukoplakia (VCL) cells.
  • Researchers used immunohistochemistry to analyze the presence of pepsin and H/K ATPase subunits in VCL cells with different levels of cell abnormality.
  • The findings revealed that higher levels of pepsin and H/K ATPase were linked to greater dysplasia in VCL, with acidified pepsin increasing cell proliferation and autophagy, which were modulated by certain inhibitors.

Article Synopsis
  • Thyroglossal duct cysts (TDCs) are usually single cysts, but this study describes an unusual case with five cysts, which is the first of its kind reported in the front neck area.
  • The treatment involved a Sistrunk operation for complete excision, and histological analysis confirmed the presence of TDCs with no recurrence after 6 years.
  • The case highlights the rarity of multiple TDCs and emphasizes the need for thorough preoperative imaging and careful assessment to avoid misdiagnosis.
