Unlabelled: Type I interferons exert their antiviral effects against SARS-CoV-2 by inducing the expression of interferon-stimulated genes (ISGs), including but not limited to LY6E, CH25H, IFITM2/3, and IFIH1. However, the antiviral effect and underlying mechanisms of action of most ISGs in SARS-CoV-2 infection are not yet fully understood. By screening 109 ISG-knockout cell lines, we identify that phospholipid scramblase 1 (PLSCR1), an interferon-inducible protein, acts as a crucial restriction factor against SARS-CoV-2 infection.

Quantitative insertion of functionality into polymer chains via step-growth polymerization is challenging due to the random bimolecular reaction and relatively low extent of reaction. Herein, a copper-catalyzed azide-alkyne cycloaddition (CuAAC) step-growth polymerization with reaction-enhanced reactivity of intermediates (RERI) mechanism is applied for the precise insertion of degradable functions into the mainchain semifluorinated polymer and endowed it with controlled degradability. In this CuAAC polymerization, bis-alkynyl-terminated (A) monomer can be quantitatively consumed when the slightly excess of 2,2-bis(azidomethyl)propane-1,3-diyl bis(2-methylpropanoate) (BiAz, B) monomer with RERI effect is employed.

The wavelet transform (WT) has gained significant attention for its ability to identify damage details within strain modes. However, edge damage in structures often remains obscured and unrecognizable when WT is applied, primarily due to edge effects. Intelligent algorithms used to assess structural damage severity often face challenges such as premature convergence and a tendency to settle on local optima.

The influenza A virus evades the host innate immune response to establish infection by inhibiting RIG-I activation through its nonstructural protein 1 (NS1). Here, we reported that receptor-transporting protein 4 (RTP4), an interferon-stimulated gene (ISG), targets NS1 to inhibit influenza A virus infection. Depletion of RTP4 significantly increased influenza A virus multiplication, while NS1-deficient viruses were unaffected.

Maintaining genomic stability is vital for cellular equilibrium. In this study, we combined CRISPR-mediated base editing with pooled screening technologies to identify numerous mutations in lysine residues and protein-coding genes. The loss of these lysine residues and genes resulted in either sensitivity or resistance to DNA-damaging agents.

Article Synopsis
  • - Tertiary lymphoid structures (TLS) are linked to various diseases, such as cancers and chronic infections, but their role in immune responses and formation processes is not completely understood.
  • - The dental pulp, with its unique immune cell organization and connection to the outside environment, provides a valuable model for studying TLS and immune responses to bacterial infections.
  • - Single-cell RNA sequencing and immunofluorescence staining in healthy versus inflamed dental pulp reveal markers and increased immune cell activity, indicating that TLS forms in inflamed conditions and may be driven by molecules like CCL3.

Anisotropic nanoparticles such as worm-like micelles have aroused much attention due to their promising applications from templates to drug delivery. The fabrication of worm-like micelles with tunable structural stability and control over their diameter and length is of great importance but still challenging. Herein, we report a kinetically controlled ring-opening metathesis polymerization-induced self-assembly (ROMPISA) for the robust preparation of kinetically trapped worm-like micelles with tunable diameter/length at enlarged experimental windows by the rational manipulation of kinetic factors, including solvent property, temperature, and π-π stacking effects.

In subsurface biodegraded oil reservoirs, methanogenic biodegradation of crude oil is a common process. This process was previously assigned to the syntrophy of hydrocarbon-degrading bacteria and methanogenic archaea. Recent studies showed that archaea of the Candidatus Methanoliparum named as alkylotrophic methanogens couple hydrocarbon degradation and methane production in a single archaeon.

CRISPR screens have empowered the high-throughput dissection of gene functions; however, more explicit genetic elements, such as codons of amino acids, require thorough interrogation. Here, we establish a CRISPR strategy for unbiasedly probing functional amino acid residues at the genome scale. By coupling adenine base editors and barcoded sgRNAs, we target 215,689 out of 611,267 (35%) lysine codons, involving 85% of the total protein-coding genes.

is a shrubby genus of about eight species distributed basically in East Asian evergreen broadleaved forests (EBLFs), with distribution centers in the karst regions of Yunnan, Guizhou, and Guangxi in southwestern China. Based on the hemiparasitic and more or less liana habits of this genus, we hypothesized that its evolution and distribution were shaped by the development of EBLFs there. To test our hypothesis, the most comprehensive phylogenies of hitherto were constructed based on plastome and nuclear loci (nrDNA, PHYA and PHYB); then divergence time and ancestral areas were inferred using the combined nuclear loci dataset.

Background: The acute exacerbation of chronic obstructive pulmonary disease (AECOPD) is a common respiratory disease among older adults, which imposes a significant burden on individuals and society and poses a major challenge to the global public health system due to its high morbidity and mortality. Acupuncture is effective for AECOPD, but its efficacy has been questioned due to the limited methodological quality. Thus, we aim to investigate the efficacy of acupuncture as adjunctive therapy for AECOPD and determine whether the efficacy of acupuncture differs with the type of acupoint combinations.

Multi-view action recognition aims to identify action categories from given clues. Existing studies ignore the negative influences of fuzzy views between view and action in disentangling, commonly arising the mistaken recognition results. To this end, we regard the observed image as the composition of the view and action components, and give full play to the advantages of multiple views via the adaptive cooperative representation among these two components, forming a Dual-Recommendation Disentanglement Network (DRDN) for multi-view action recognition.
