To better understand human health and disease, researchers create a wide variety of mouse models that carry human DNA. With recent advances in genome engineering, the targeted replacement of mouse genomic regions with orthologous human sequences has become increasingly viable, ranging from finely tuned humanisation of individual nucleotides and amino acids to the incorporation of many megabases of human DNA. Here, we examine emerging technologies for targeted genomic humanisation, we review the spectrum of existing genomically humanised mouse models and the insights such models have provided, and consider the lessons learned for designing such models in the future.

Introduction: Although mutations in the human beta-globin gene cluster are essentially point mutations, several large deletions have been described in recent years.

Methods: We have identified a novel 223 kb deletion in a Chinese patient by multiplex ligation-dependent probe amplification and characterized it by next-generation sequencing, Gap-PCR, and DNA sequence analysis.

Results: The deletion extends from the 3'UTR of the δ globin gene (HBD) to 215 kb downstream of the HBB.

Bending straight Ag nanorods (AgNRs) into V-shaped structures can generate a higher surface-enhanced Raman scattering (SERS) performance. Numerical simulations showed that V-shaped AgNRs with a total length between 300 nm and 800 nm were more sensitive than equal-length straight AgNRs under a 785 nm laser in most cases. It was found that at a laser wavelength between 500 nm and 1000 nm, the Raman enhancement factor (EF) of a V-shaped AgNR's 3rd plasmon mode was not only optimal among the other major plasmon modes, but also outperformed the plasmon modes of straight AgNRs.

Based on the data of 1179 discs and whorls of 49 trees from larch (Larix olgensis) plantations located in Mengjiagang forest farm in Heilongjiang Province, China, we analyzed the longitudinal variation pattern of heartwood radius. The results showed that the heartwood radius decreased with the increases of tree height, which was basically the same as the trunk shape. The relationship between the xylem radius (XR), diameter at breast height (DBH) and cambial age (CA) with the heartwood radius was significant.

Inspired by glucose-sensitive ion channels, herein we describe a biomimetic glucose-enantiomer-driven ion gate via the introduction of the chiral pillar[6]arene-based host-guest systems into the artificial nanochannels. The chiral nanochannels show a high chiral-driven ionic gate for glucose enantiomers and can be switched "off" by D-glucose and be switched "on" by L-glucose. Remarkably, the chiral nanochannel also exhibited a good reversibility toward glucose enantiomers.

Despite career construction theory attends to individual subjective career and provides a useful lens to study well-being, extant research has yielded limited insights into the mechanisms through which career construction variables influence individual well-being. To address this important gap, the present study examined a mediation model that links indicators of career adaptivity (big-five personality and approach/avoidance traits) to psychological well-being (psychological flourishing and life satisfaction) through career adaptability and in sequent meaning of life (presence of life meaning and search for life meaning) among a sample of Chinese university students ( = 165). The results of a two-wave survey study showed that career adaptability and presence of life meaning mediated the effects of openness to experience, consciousness, approach trait, and avoidance trait on individual well-being in sequence.
