Cell surface receptor-targeted protein degraders hold promise for drug discovery. However, their application is restricted because of the complexity of creating bifunctional degraders and the reliance on specific lysosome-shuttling receptors or E3 ubiquitin ligases. To address these limitations, we developed an autophagy-based plasma membrane protein degradation platform, which we term AUTABs (autophagy-inducing antibodies).

Purpose: Camrelizumab and rivoceranib together provide a new first-line treatment approach for unresectable hepatocellular carcinoma (HCC). Meanwhile, transarterial chemoembolization (TACE) is an effective method for the local control of the HCC. The study compared the clinical benefit and safety between TACE with camrelizumab-rivoceranib and camrelizumab-rivoceranib alone for Barcelona Clinic Liver Cancer (BCLC)-C HCC patients.

Objective: To evaluate the baseline albumin-bilirubin (ALBI) grade's role in advanced hepatocellular carcinoma (HCC) receiving transarterial chemoembolization (TACE) plus anti-angiogenesis therapies and PD-1 inhibitors (TACE+TP) versus anti-angiogenesis therapies and PD-1 inhibitors (TP).

Methods: This multicenter retrospective study enrolled advanced HCC undergoing TACE+TP or TP from January 2019 to June 2023 at three hospitals in China. The primary outcomes were time to progression of the ALBI grade and change in ALBI score between the initial baseline and the final assessment point available, the secondary outcomes consisted of overall survival (OS) as well as progression-free survival (PFS).

Article Synopsis
  • - Fumarate hydratase (FH) deficiency is linked to hereditary leiomyomatosis and renal cell carcinoma (RCC), and FH-deficient tumors currently have limited treatment options.
  • - Researchers used an epigenetic-focused guide RNA library to identify the chromodomain helicase DNA binding protein 6 (CHD6) as a key factor promoting the growth of FH-mutated RCC through its interaction with pro-inflammatory enhancers and NF-κB-related transcription.
  • - The study found that a specific degrader (AU-15330) could effectively target CHD6 and inhibit the growth of FH-deficient RCC tumors, revealing potential new therapeutic strategies for these cancers.

Temozolomide (TMZ) serves as the principal chemotherapeutic agent for glioma; nonetheless, its therapeutic efficacy is compromised by the rapid emergence of drug resistance, the inadequate targeting of glioma cells, and significant systemic toxicity. ARV-825 may play a role in modulating drug resistance by degrading the BRD4 protein, thereby exerting anti-glioma effects. Therefore, to surmount TMZ resistance and achieve efficient and specific drug delivery, a dual-targeted glutathione (GSH)-responsive nanoparticle system (T+A@Glu-NP) is designed and synthesized for the co-delivery of ARV-825 and TMZ.

  • Early pregnancy features high levels of natural killer (NK) cells at the maternal-fetal interface, which are crucial for fetal development and remodeling uterine arteries.
  • Aberrant activity of decidual NK (dNK) cells is linked to recurrent pregnancy loss (RPL), but the exact molecular mechanisms behind their function need more research.
  • This study highlights that downregulation of miR-122-5p in dNK cells from RPL patients affects their function, suggesting that miR-122-5p may play a role in maintaining immune tolerance during pregnancy.

Background: The effectiveness of the anastomosis method for laparoscopic left colectomy (LLC) remains inconclusive. Thus, a systematic review and meta-analysis were conducted to compare the outcomes between intraperitoneal anastomosis (IPA) and extraperitoneal anastomosis(EPA)in LLC.

Methods: PubMed, Embase, the Cochrane Library, CNKI, and WanFangData were systematically searched for relevant literature.

Assessing and selecting climate models with lower uncertainty is necessary to predict future climate and hydrological risks at the watershed scale. In this study, we integrated stepwise cluster analysis (SCA) to propose a multi-model ensemble downscaling framework aimed at reducing the uncertainty of GCM-based precipitation projections in large-scale watersheds. The Pearl River Basin (PRB) in southern China was selected as the study area to validate the reliability of this framework.

Programming the organization of discrete building blocks into periodic and quasi-periodic arrays is challenging. Methods for organizing materials are particularly important at the nanoscale, where the time required for organization processes is practically manageable in experiments, and the resulting structures are of interest for applications spanning catalysis, optics, and plasmonics. While the assembly of isotropic nanoscale objects has been extensively studied and described by empirical design rules, recent synthetic advances have allowed anisotropy to be programmed into macroscopic assemblies made from nanoscale building blocks, opening new opportunities to engineer periodic materials and even quasicrystals with unnatural properties.

Background: This study aims to assess the associations of admission systolic blood pressure (SBP) level with spontaneous reperfusion (SR) and long-term prognosis in ST-elevation myocardial infarction (STEMI) patients.

Methods: Data from 3809 STEMI patients who underwent primary percutaneous coronary intervention within 24 h, as recorded in the Chinese STEMI PPCI Registry (NCT04996901), were analyzed. The primary endpoint was SR, defined as thrombolysis in myocardial infarction grade 2-3 flow of IRA according to emergency angiography.

  • The endometrium transforms into a structure called decidua during pregnancy preparation, with senescent decidual stromal cells (DSCs) increasing but poorly understood mechanisms.
  • Researchers used single-cell RNA transcriptomics to discover that higher levels of the BCAA transporter SLC3A2 accompany cell senescence in DSCs during decidualization.
  • A high leucine diet contributes to increased DSC senescence and embryo loss in mice, while dNK cells can counteract this senescence through the TNFSF14/TNFRSF14 pathway, highlighting potential for therapeutic interventions in preventing pregnancy loss.

Colloidal crystal engineering with DNA allows one to design diverse superlattices with tunable lattice symmetry, composition, and spacing. Most of these structures follow the complementary contact model, maximizing DNA hybridization on building blocks and producing relatively close-packed lattices. Here, low-symmetry kagome superlattices are assembled from DNA-modified gold bipyramids that can engage only in partial DNA surface matching.

As the thickness of an ultra-thin flattened heat pipe (UTHP) decreases, the fabrication difficulty increases exponentially, and the thermal performance deteriorates rapidly. In this study, three types of composite wicks were developed for UTHPs with a 0.6 mm thickness: copper foam and mesh wick (CFMW), two layers of different mesh wick (TDMW), and three layers of the same mesh wick (TSMW).

  • RBC counts decrease with age, but this trend is significant in men only, while women do not show the same pattern.
  • The study utilized various methods to analyze the presence and uptake of mitochondrial DNA (mtDNA) in red blood cells (RBCs) from both young and older adults.
  • Elevated mtDNA levels in RBCs are linked to increased oxidative stress, suggesting that mtDNA content could indicate RBC aging, with men showing more rapid senescence than women.

The purpose of this study was to explore whether prenatal gut microbiota (GM) and its functions predict the development of offspring temperament. A total of 53 mothers with a 1-year-old child and 41 mothers with a 2-year-old child were included in this study using a mother-infant cohort from central China. Maternal fecal samples collected during the third trimester were analyzed using 16S rRNA V3-V4 gene sequences.
