DNA replication, a fundamental process in all living organisms, proceeds with continuous synthesis of the leading strand by DNA polymerase ε (Pol ε) and discontinuous synthesis of the lagging strand by polymerase δ (Pol δ). This inherent asymmetry at each replication fork necessitates the development of methods to distinguish between these two nascent strands in vivo. Over the past decade, strand-specific sequencing strategies, such as enrichment and sequencing of protein-associated nascent DNA (eSPAN) and Okazaki fragment sequencing (OK-seq), have become essential tools for studying chromatin replication in eukaryotic cells.

Cancer vaccines hold great promise in the fight against cancer. Here, we report an ice-pop making inspired photothermal ultra-swelling microneedle (PUSMN) patch for facilitating and enhancing cancer vaccination. The PUSMN patch consist of an array of microneedles made from photo-crosslinked methacrylated hyaluronic acid and polydopamine, a near-infrared photothermal conversion material, connected to a customized resin handle like an ice-pop stick.

Rationale And Objectives: To develop and validate a radiomics signature, utilizing baseline and restaging CT, for preoperatively predicting progression-free survival (PFS) after neoadjuvant chemotherapy (NAC) in locally advanced gastric cancer (LAGC).

Methods: A total of 316 patients with LAGC who received NAC followed by gastrectomy were retrospectively included in this single-center study; these patients were split into two cohorts, one for training (n = 243) and the other for validation (n = 73), based on the different districts of our hospital. A total of 1316 radiomics features were extracted from the volume of interest of the gastric-cancer lesion on venous phase CT images.

Background: Amulirafusp alfa (IMM0306) is a fusion protein of CD47 binding domain of signal-regulatory protein alpha (SIRPα) with CD20 monoclonal antibody on both heavy chains. This study aimed to evaluate the safety and preliminary efficacy of amulirafusp alfa in relapsed or refractory (r/r) B-cell non-Hodgkin lymphoma (B-NHL).

Methods: We enrolled patients with CD20 + r/r B-NHL who had previously received at least two lines of therapy to receive a single-dose of amulirafusp alfa in the first 2 weeks, followed by a multiple-dose period, in which the patients received the same intravenous dose every week in 4-week cycles.

The identification of ferroptosis-sensitive cancers is critical for the application of ferroptosis-inducing therapies in cancer therapy. Here, patient-derived organoid screening models of colorectal cancer are established to identify tumors that are sensitive to ferroptosis-inducing therapy. This study discovers that patient-derived tumors characterized by mitophagy deficiency are hypersensitive to ferroptosis-inducing therapies.

In clinical trials, we often encounter observations from patients' paired organs. In paired correlated data, there exist various measures to evaluate the therapeutic responses, such as risk difference, relative risk ratio, and odds ratio. These measures are essentially some forms of response rate functions.

In the paper, metabolomics techniques based on UHPLC-QE-MS were used to study raw black beans, steaming black beans, and their in vitro digestion products. The results show that the three groups of raw black beans, atmospheric pressure-steamed black beans, and their in vitro digests comprised 922, 945, and 878 characteristic metabolites, respectively, dominated by amino acids, organic acids, polyphenols, and sugars. After screening the differential metabolites, content comparison, the content of amino acids, sugars, and phenolics in black beans was found to be increased after atmospheric steaming.

Soft-magnetic fibers (SMFs) play a crucial role in energy conversion, transmission, and storage within electronic devices. However, conventional SMFs have poor plasticity and are therefore difficult to withstand long-term tensile, torsional, and shear deformation. A high fraction of grain boundaries could improve plastic deformability of conventional SMFs, but deteriorates the coercivity.

Methotrexate (MTX) is a primary treatment for moderate to severe psoriasis, but conventional oral or subcutaneous delivery has limitations, including poor drug targeting, side effects, and low patient compliance. This study introduces implantable hydrogel microneedles (HMNs) made from photo-cross-linked gelatin methacryloyl (GelMA) and puerarin (Pue), a traditional Chinese medicine, for intradermal and sustained MTX delivery. The ratio of Pue to GelMA significantly influenced the morphology, mechanical strength, MTX release profiles, and degradation behavior of the HMNs.

Objective: Explore alterations in topological features of gray matter volume (GMV) and structural networks in non-cognitive impairment end-stage renal disease (Non-CI ESRD).

Materials And Methods: Utilizing graph theory, we collected structural magnetic resonance imaging (sMRI) data from 38 Non-CI ESRD patients and 50 normal controls (NC). We compared, and extracted the GMV across subject groups, constructed corresponding structural covariance networks (SCNs), and investigated the alterations in SCNs feature parameters between groups.

Article Synopsis
  • Superfine cement is favored in construction and environmental projects for its strength and durability but has issues like high bleeding rates and long setting times.
  • This study tests new expansive superfine cement slurries with added agents to improve fluidity, reduce setting times, and enhance strength, showing significant performance improvements over traditional formulas.
  • Results indicate that the optimal mix proportions lead to faster setting times and higher compressive strengths, along with improved volume expansion rates, enhancing the material's effectiveness in practical applications.

Soft magnetic materials (SMMs) are indispensable for electromechanical energy conversion in high-efficiency applications, but they are exposed to increasing mechanical loading conditions in electric motors due to higher rotational speeds. Enhancing the yield strength of SMMs is essential to prevent the degradation in magnetic performance and failure from plastic deformation, yet most SMMs have yield strengths far below one gigapascal. Here, we present a multicomponent nanostructuring strategy that doubles the yield strength of SMMs while maintaining ductility.

1,2-Dichloroethane (1,2-DCE), a prevalent industrial and environmental contaminant, induces toxic encephalopathy through inhalation, leading to neurotoxic effects and inflammation-driven brain edema. Human umbilical cord mesenchymal stem cells (HUCMSCs) secrete bioactive factors, including miRNAs, proteins, and lipids via exosomes, exhibiting anti-inflammatory and immune-regulatory properties. However, their potential in treating 1,2-DCE-induced neuroinflammation and the underlying mechanisms remain unclear.

Objective: Intracytoplasmic sperm injection (ICSI) is commonly employed in preimplantation genetic testing (PGT) to minimize the risk of foreign sperm DNA contamination. Cryopreserved embryos from patients with recurrent miscarriage or repeated implantation failure, who have undergone conventional fertilization (IVF), can be thawed and biopsied for PGT. Therefore, we aimed to assess the accuracy and effectiveness of preimplantation genetic testing for aneuploidy (PGT-A) on frozen embryos using conventional IVF (c-IVF) insemination methods.

Article Synopsis
  • Some follicular lymphoma (FL) patients on a "watch and wait" strategy experience quick disease progression, prompting researchers to find ways to identify these individuals and develop a risk score for better screening at diagnosis.
  • In a study involving 411 FL patients from 16 cancer centers, 35% required treatment within 24 months, with a significantly lower 5-year progression-free survival rate (62.3% vs. 89.5%) for those needing treatment early.
  • Five key factors were identified as predictors for needing treatment within 24 months: advanced disease stages, high β microglobulin levels, low lymphocyte-to-monocyte ratios, bone marrow involvement, and spleen enlargement; these factors can help pinpoint patients who
