Objective: To investigate DNA methylation (DNAm) status of dickkopf-associated protein 1 (DKK-1) in ossified hip capsule synovium and serum among patients with ankylosing spondylitis (AS).

Methods: Western blot was applied to detect the level of DKK-1 protein expression in hip joint capsule tissues from four patients with AS as well as four patients with femoral neck fracture (FNF) caused by trauma as control. DKK-1 gene promoter methylation (GPM) was examined by methylation-specific polymerase chain reaction.

Article Synopsis
  • - The study focuses on a newly isolated bacterium, Pseudomonas aeruginosa YL, which demonstrates effective simultaneous nitrification and denitrification, addressing the inefficiencies of traditional methods.
  • - Key optimal conditions for nitrogen removal were identified: succinate as a carbon source, a C/N ratio of 10, pH 7.0, a temperature of 30°C, and a shaking speed of 160-200 r/min, achieving high removal rates of ammonia, nitrogen, and TOC.
  • - Under these optimal conditions, strain YL achieved impressive nitrate and total nitrogen removal rates of 94.6% and 76.3%, highlighting its potential for efficient nitrogen removal in wastewater treatment.

Article Synopsis
  • * The strain preferentially removed ammonium when mixed nitrogen sources were present, and specific genes related to these processes were successfully amplified, indicating biological activity.
  • * Optimal conditions for nitrite removal were identified, with dissolved oxygen being the key factor, and enriched bacteria in a sequencing batch reactor led to stable nitrogen and organic removal, showing promise for practical applications.
