Aryldihydronaphthalenes (ADHNs) and their derivatives are widely found in many types of natural products, bioactive compounds, and functional materials, and are also important synthetic intermediates in organic chemistry, attracting widespread attention from both organic and pharmaceutical chemists. In the past two decades, the chemical synthesis and biological activity of ADHNs and their derivatives have become two hot spots. This review summarizes the synthetic protocols of ADHN derivatives, introduces some representative examples of the reaction mechanism, and focuses on the research progress of ADHNs in natural product chemistry and chemical biology since 2000.

A series of novel 2-substituted 2, 3-dihydroquinazolin-4(1H)-one derivatives were designed, synthesized and estimated for their in vitro antiproliferative activities against HepG2, U251, PANC-1, A549 and A375 cell lines. Among them, compound 32 was the most promising candidate, and displayed strong broad-spectrum anticancer activity. The mechanism studies revealed that compound 32 inhibited tubulin polymerization in vitro, disrupted cell microtubule networks, arrested the cell cycle at G2/M phase, and induced apoptosis by up-regulating the expression of cleaved PARP-1 and caspase-3.

In order to provide corresponding anesthesia methods of hepatobiliary diseases and obtain better clinical effect, the hemodynamics and clinical efficacy are evaluated by using different doses of dexamethasone (DEX) during the operation. 97 patients undergoing general anesthesia for hepatobiliary surgery in our hospital from April 2020 to August 2021 are analyzed retrospectively. All patients are divided into G1 ( = 24), G2 ( = 24), G3 ( = 24), and control group ( = 25) according to the random number table method.
