The Streptococcus canis Cas9 protein (ScCas9) recognizes the NNG protospacer adjacent motif (PAM), offering a wider range of targets than that offered by the commonly used S. pyogenes Cas9 protein (SpCas9). However, both ScCas9 and its evolved Sc++ variant still exhibit low genome editing efficiency in plants, particularly at the less preferred NTG and NCG PAM targets.

A three-dimensional solid model of foot drop was constructed using anatomical structures segmented from medical CT scans, including bone, cartilage, ligaments, plantar fascia, and soft tissues, and the impact velocities of the foot were set to be 2 m/s, 4 m/s, 6 m/s, 8 m/s, and 10 m/s. Explicit kinetic analysis methods were used to investigate the mechanical response of the foot landing with different speeds to explore the damage mechanism of heel bone at different impact velocities.

Methods: Using conical beam CT (CBCT) images of a healthy person, models of the complete maxillary anterior dental region (Model A) and maxillary anterior dental region with a missing left maxillary central incisor (Model B) were constructed. The labial and palatine alveolar bone and tooth surface of the bilateral incisor and cusp regions were selected as the application sites, the resting perioral force was applied perpendicular to the tissue surface, and the changes in maxillary stress and displacement after the perioral force was simulated were analyzed.
Introduction: Preclinical studies have shown that oxygen therapy can improve ischaemic brain tissue oxygen tension, reduce reperfusion injury after revascularisation, promote neuroregeneration and inhibit inflammatory responses potentially exerting a beneficial effect after endovascular treatment (EVT) in patients with acute ischaemic stroke (AIS). However, the optimal fraction of inspired oxygen (FiO) during EVT under general anaesthesia is currently unknown. Therefore, we are conducting a randomised controlled trial (RCT) to evaluate the impact of high-concentration oxygen vs low-concentration normobaric oxygen on early neurological function after EVT.

Purpose: Liver disease is associated with a range of extrahepatic complications, which have recently been expanded to include ophthalmic conditions. However, evidence is lacking regarding its impact on primary open-angle glaucoma (POAG). This study aimed to investigate whether major liver diseases, including metabolic dysfunction-associated steatotic liver disease (MASLD), alcoholic liver disease (ALD), viral hepatitis, and liver fibrosis and cirrhosis, were associated with POAG.

In this work, the degradation mechanisms, kinetics, and ecotoxicity of kaempferol and quercetin mediated by ∙OH, ∙OCH, ∙OOH, and O in gaseous and aqueous environments were investigated using cell experiments and quantum chemical calculations. Three radical scavenging mechanisms, including hydrogen atom transfer (HAT), radical adduct formation (RAF) and single electron transfer (SET) were discussed.

Objectives: In the present study with a large cohort, we aimed to characterize intracerebral seizure onset patterns (SOP) of mesial temporal lobe epilepsy (mTLE), with or without hippocampal sclerosis (HS) as identified via magnetic resonance imaging (MRI).
Methods: We retrospectively analyzed 255 seizures of 76 consecutive patients with mTLE explored by stereoelectroencephalography (SEEG), including HS-mTLE (n = 52) and non-HS- mTLE (n = 24). Relevant results were obtained by a combination of spectral analysis and manual review.
Quaternary ammonium salts (QAs) are a class of highly active compounds widely used in medicine and agriculture. However, many QAs lack a conjugated system, making their recognition and quantitation challenging. Stachydrine is a representative unconjugated QA with a high content in Houtt.

Purpose: To investigate possible associations between inaccurate femoral tunnel placement during MPFL-R and clinical outcomes, with a specific focus on proximal tunnel malpositioning.
Study Design: Cohort study; Level of evidence, 3.
CDK4/6i, the first-line drug for treating ERα-positive breast cancer, significantly improves clinical outcomes. However, CDK4/6i resistance often develops and remains a major hurdle, and the underlying mechanisms remain challenging to fully investigate. Here, we used Genome-wide CRISPR/Cas9 library screening combined with single-cell sequencing to screen for molecules mediating CDK4/6i resistance and identified METTL14 as a determinant of CDK4/6i sensitivity.

This communication is important in thermogenic adipocytes to increase the energy expenditure that controls body temperature and weight. However, the regulatory mechanisms of OMM protein insertion are poorly understood.

Despite growing consumer concerns about water flavor, a spatial assessment of the determinants of water flavor in China has been notably lacking. This study bridges this gap by conducting a spatially comprehensive analysis of 78 tap water samples throughout China.

Argonaute 2 (Ago2) is a crucial enzyme in the RNA interference (RNAi) pathway, essential for gene silencing via the cleavage of target messenger RNA (mRNA) mediated by microRNA (miRNA) or small interfering RNA (siRNA). The activity of Ago2 is a significant biomarker for various diseases, including cancer and viral infections, necessitating precise monitoring techniques. Traditional methods for detecting Ago2 activity are often cumbersome and lack the necessary sensitivity and specificity for low-abundance targets in complex samples.

Disentangling the contributions from fluctuations in the nuclear overlap size (geometrical component) and other sources at a fixed size (intrinsic component) remains a challenge. This problem is addressed by measuring the mean, variance, and skewness of P([p_{T}]) in ^{208}Pb+^{208}Pb and ^{129}Xe+^{129}Xe collisions at sqrt[s_{NN}]=5.

Nonetheless, a limitation of many existing flexible force sensors stems from their isotropic structure or material properties, preventing them from simultaneously detecting both the direction and magnitude of the applied force. Herein, a high-performance 3D force sensor based on orthogonal multimodal sensing, the cancellation principle, and the strain effect is proposed.

As a recent focal point of research, soft electronics encompass various factors that synergistically enhance their mechanical properties and ensure stable electrical performance. However, challenges such as immiscible conductive fillers, poor phase interfaces, and unstable conductive networks hinder the overall efficacy of these materials. To address these issues, a hydrogel featuring an oriented interpenetrating network structure (OIPN) is developed.
