Hereditary spastic paraplegia (HSP) is a group of neurodegenerative diseases with genetic and clinical heterogeneity characterized by spasticity and weakness of the lower limbs. It includes four genetic inheritance forms: autosomal dominant inheritance (AD), autosomal recessive inheritance (AR), X-linked inheritance, and mitochondrial inheritance. To date, more than 82 gene loci have been found to cause HSP, and SPG15 () is one of the most common autosomal recessive hereditary spastic paraplegias (ARHSPs) with a thin corpus callosum (TCC), presents with early cognitive impairment and slowly progressive leg weakness.

Article Synopsis
  • A meta-analysis was conducted to determine the relationship between angiotensin converting enzyme (ACE) gene polymorphisms and Alzheimer's disease (AD) risk, using data from 82 cohorts across 65 studies.
  • While the study identified some associations, particularly with the rs1799752 polymorphism suggesting the insertion allele (I) might increase the risk of AD, these results were not robust after statistical adjustments and did not hold consistently across different populations.
  • Ultimately, the findings indicate mixed results, with significant associations found in late-onset AD and in North Europeans, but overall there was no definitive link after controlling for genetic equilibrium.

The third-generation of epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs), represented by osimertinib, has achieved remarkable clinical outcomes in the treatment of non-small-cell lung cancer (NSCLC) with EGFR mutation. However, resistance eventually emerges in most patients and the underlying molecular mechanisms remain to be fully understood. In this study, we generated an osimertinib-acquired resistant lung cancer model from a NSCLC cell line H1975 harboring EGFR L858R and T790M mutations.

Article Synopsis
  • 5-aza-2'-deoxycytidine (5-aza-CdR) is an anticancer drug that can reactivate the tumor suppressor gene CDX2 in gastric cancer, leading to improved survival rates in patients expressing CDX2.
  • In gastric cancer tissues, higher levels of DNA methyltransferase 1 (DNMT1) correlate with advanced disease features, while CDX2 expression is inversely related to DNMT1, suggesting that DNMT1 silences CDX2.
  • Treatment with 5-aza-CdR demethylates the CDX2 promoter, increases its expression, reduces DNMT1 levels, and leads to inhibited cell growth and increased apoptosis in gastric cancer cell

Anisodamine, which is originally extracted from scopolia tangutica and is currently produced in China, is a tropane alkaloid and a muscarinic cholinoceptor blocker. Our previous study found that anisodamine did not alter high K(+)-evoked contraction of rabbit aortic rings using isometric tension recording methods, but could attenuate noradrenaline (NA)-, histamine- or 5-hydroxytryptamine-induced contraction in an endothelium-independent manner. Since the high K(+)-elicited depolarization non-selectively inhibits potassium channels in vascular smooth muscle cell (VSMC) membrane, the vasodilation effect of some potassium channel activators may be inhibited or abolished in high K(+) solution.
