Histone post-translational modifications (PTMs) regulate gene expression patterns through epigenetic mechanisms. The 5 histone proteins (H1, H2A, H2B, H3, and H4) are extensively modified, with over 75 distinct modification types spanning more than 200 sites. Despite strong advances in mass spectrometry-based approaches, identification and quantification of modified histone peptides remains challenging due to factors such as isobaric peptides, pseudo-isobaric PTMs, and low stoichiometry of certain marks.

Background: Covered stent correction for a sinus venosus atrial septal defect (SVASD) was first performed in 2009. This innovative approach was initially viewed as experimental and was reserved for highly selected patients with unusual anatomic variants. In 2016, increasing numbers of procedures began to be performed, and in several centers, it is now offered as a standard of care option alongside surgical repair.

Reversible modification of the histone H3 N-terminal tail is critical in regulating the chromatin structure, gene expression, and cell states, while its dysregulation contributes to disease pathogenesis. Understanding the crosstalk between H3 tail modifications in nucleosomes constitutes a central challenge in epigenetics. Here, we describe an engineered sortase transpeptidase, cW11, that displays highly favorable properties for introducing scarless H3 tails onto nucleosomes.

This consensus document for the performance of cardiovascular computed tomography (CCT) to guide intervention in the right ventricular outflow tract (RVOT) in patients with congenital disease (CHD) was developed collaboratively by pediatric and adult interventionalists, surgeons and cardiac imagers with expertise specific to this patient subset. The document summarizes definitions of RVOT dysfunction as assessed by multi-modality imaging techniques and reviews existing consensus statements and guideline documents pertaining to indications for intervention. In the context of this background information, recommendations for CCT scan acquisition and a standardized approach for reporting prior to surgical or transcatheter pulmonary valve replacement are proposed and presented.

Dysregulated transcription due to disruption in histone lysine methylation dynamics is an established contributor to tumorigenesis. However, whether analogous pathologic epigenetic mechanisms act directly on the ribosome to advance oncogenesis is unclear. Here we find that trimethylation of the core ribosomal protein L40 (rpL40) at lysine 22 (rpL40K22me3) by the lysine methyltransferase SMYD5 regulates mRNA translation output to promote malignant progression of gastric adenocarcinoma (GAC) with lethal peritoneal ascites.

This work presents a novel and versatile approach to employ textile capacitive sensing as an effective solution for capturing human body movement through fashionable and everyday-life garments. Conductive textile patches are utilized for sensing the movement, working without the need for strain or direct body contact, wherefore the patches can sense only from their deformation within the garment. This principle allows the sensing area to be decoupled from the wearer's body for improved wearing comfort and more pleasant integration.

Article Synopsis
  • Protein arginylation is a crucial posttranslational modification done by the enzyme ATE1, which involves adding an arginine to proteins, making it hard to distinguish from normal arginine present in proteins.
  • Researchers introduced a new method called activity-based arginylation profiling (ABAP) that uses isotopic labeling to identify arginylation in various biological samples without interference from translational activities.
  • ABAP successfully identified 229 unique arginylation sites in human proteins and can be applied to different samples, helping advance the understanding of this complex protein modification's biological roles.

Background: Stent implantation has become standard of care in older children and adults for treatment of branch pulmonary artery stenosis (BPAS) and coarctation aorta (CoAo). There are no stents approved or available for infants that have the potential to be dilated to adult diameters. The Minima stent was designed to fulfill this unmet need.

Background: Transcatheter closure of the patent ductus arteriosus (PDA) in premature infants is currently dependent on fluoroscopic guidance and transportation to the catheterization laboratory.

Aim: We describe a new echocardiographically guided technique to allow our team to move to the bedside at the neonatal intensive care unit (NICU) of the referring center for percutaneous treatment of PDA in premature infants.

Methods: This is a single-center, retrospective, primarily descriptive analysis.

The interest in MS-based analysis of modified nucleic acids is increasing due to the application of nucleic acids in therapeutics. However, there are few available integrated platforms for characterizing nucleic acid modifications. Herein, we report a general mass spectrometry-based SWATH platform to identify and quantify both RNA and DNA modifications, which we call SWATH analysis of modified nucleic acids (SWAMNA).

Introduction: Clinical pharmacists are well positioned to enhance efforts to promote emergency department (ED)-initiated buprenorphine to treat opioid use disorder (OUD). Among clinical pharmacists in urban EDs, we sought to characterize barriers and facilitators for ED-initiated buprenorphine to inform future implementation efforts and enhance access to this highly effective OUD treatment.

Methods: This study was conducted as a part of Project ED Health (CTN-0069, NCT03023930), a multisite effectiveness-implementation study aimed at promoting ED-initiated buprenorphine that was conducted between April 2017 and July 2020.

  • SPINDLY (SPY) is a newly discovered nucleocytoplasmic protein O-fucosyltransferase (POFUT) in Arabidopsis thaliana that plays a crucial role in various developmental processes.
  • The structure of SPY, determined through cryo-electron microscopy, shows it uniquely binds GDP-fucose instead of the typical UDP-GlcNAc, and it forms an antiparallel dimer unlike human counterparts.
  • The N-terminal peptide of SPY contains self-fucosylation sites that inhibit its activity, while specific regions (TPRs 1-5) regulate its function by affecting how it interacts with protein substrates.

Effective conservation of endangered species relies on the characterization of habitat use and tracking of long-term population trends, which can be especially challenging for marine species that migrate long distances and utilize a diversity of habitats throughout their lives. Since 2012, citizen science volunteers at the Aquarium of the Pacific in Long Beach, California, have been monitoring an urban population of East Pacific green sea turtles () that resides near the mouth of the San Gabriel River (SGR) in Southern California, USA, in order to gain insights about how the population uses this area. Here, we collate and analyze nine years of citizen science data, including observed sightings collected across 10 observation stations.

SPINDLY (SPY) is a novel nucleocytoplasmic protein O-fucosyltransferase that regulates target protein activity or stability via O-fucosylation of specific Ser/Thr residues. Previous genetic studies indicate that AtSPY regulates plant development during vegetative and reproductive growth by modulating gibberellin and cytokinin responses. AtSPY also regulates the circadian clock and plant responses to biotic and abiotic stresses.

Unlabelled: Improved accuracy of evapotranspiration (ET) estimation, including its partitioning between transpiration (T) and surface evaporation (E), is key to monitor agricultural water use in vineyards, especially to enhance water use efficiency in semi-arid regions such as California, USA. Remote-sensing methods have shown great utility in retrieving ET from surface energy balance models based on thermal infrared data. Notably, the two-source energy balance (TSEB) has been widely and robustly applied in numerous landscapes, including vineyards.

Transcatheter pulmonary valve replacement was first performed by Dr Philip Bonhoeffer, who implanted a Medtronic Melody valve in a human in 2000. Over the past 2 decades, there have been many advances in transcatheter pulmonary valve technology. This includes the use of the SAPIEN transcatheter heart valve in the pulmonary position, modifications and refinements to valve implantation procedures, and development of self-expanding valves and prestents to treat large diameter native or patched right ventricular outflow tracts.
