Decades of research have established that mammalian transcription factors (TFs) bind to each gene's regulatory regions and cooperatively control tissue specificity, timing, and intensity of gene transcription. Mapping the combination of TF binding sites genome wide is critically important for understanding functional genomics. Here, we report a technique to measure TFs' binding sites on the human genome with a near single-base resolution by footprinting with deaminase (FOODIE) on a single-molecule and single-cell basis.

  • Cardiac ischemia/reperfusion (I/R) injury is a significant concern for heart disease, but there's currently no effective treatment to minimize this type of heart damage.
  • This study recruited patients with acute myocardial infarction to examine the impact of plasma RIPK3 levels before and after treatment, using both patient data and lab experiments on cells and mouse models.
  • Elevated RIPK3 levels post-treatment were linked to worse outcomes, and experiments showed that RIPK3 worsens heart injury by triggering inflammation, suggesting its role as a damaging molecule that could be targeted for therapy.

Background And Objectives: Noninvasive and accurate biomarkers of neurologic Wilson disease (NWD), a rare inherited disorder, could reduce diagnostic error or delay. Excessive subcortical metal deposition seen on susceptibility imaging has suggested a characteristic pattern in NWD. With submillimeter spatial resolution and increased contrast, 7T susceptibility-weighted imaging (SWI) may enable better visualization of metal deposition in NWD.

Background: The high heterogeneity of tumour and the complexity of tumour microenvironment (TME) greatly impacted the tumour development and the prognosis of cancer in the era of immunotherapy. In this study, we aimed to portray the single cell-characterised landscape of lung adenocarcinoma (LUAD), and develop an integrated signature incorporating both tumour heterogeneity and TME for prognosis stratification.

Methods: Single-cell tagged reverse transcription sequencing (STRT-seq) was performed on tumour tissues and matched normal tissues from 14 patients with LUAD for immune landscape depiction and candidate key genes selection for signature construction.

Background: Approximately 60% of patients with colorectal liver metastases (CRLM) experience relapse within 2 years after radical resection, previous studies have proven that repeat local treatment (LT) could prolong survival, however, it is difficult to seize the window for LT due to the lack of a high-sensitive surveillance method. In this study, the authors aim to examine the value of longitudinal circulating tumor DNA (ctDNA) in guiding adjuvant chemotherapy, optimizing clinical surveillance strategy, and thereby improving CRLM outcomes.

Materials And Methods: The authors conducted a prospective clinical trial using a personalized, tumor-informed ctDNA assay to monitor 60 CRLM patients undergoing resection with curative intent.

Article Synopsis
  • The study aimed to assess how a history of cancer affects the benefits of intensive blood pressure control on major cardiovascular events, using data from the SPRINT trial.
  • It included 9,336 patients, with 22.2% being cancer survivors, and found that both groups had similar risks for primary cardiovascular outcomes over a median follow-up of 3.2 years.
  • The results suggest that intensive blood pressure treatment is beneficial for both cancer survivors and those without a cancer history, indicating that past cancer treatment should not prevent aggressive hypertension management.

Background: The importance of mitochondria in normal heart function are well recognized and recent studies have implicated changes in mitochondrial metabolism with some forms of heart disease. Previous studies demonstrated that knockdown of the mitochondrial ribosomal protein S5 (MRPS5) by small interfering RNA (siRNA) inhibits mitochondrial translation and thereby causes a mitonuclear protein imbalance. Therefore, we decided to examine the effects of MRPS5 loss and the role of these processes on cardiomyocyte proliferation.
