Article Synopsis
  • Primary familial brain calcification (PFBC) is a genetic neurological disorder with no current effective treatment, linked to mutations in the SLC20A2 gene.
  • Researchers identified five new genetic variants in the SLC20A2 gene that disrupt normal splicing of its pre-mRNA, leading to dysfunctional protein production.
  • The use of splice-switching antisense oligonucleotides (ASOs) not only helped restore functional SLC20A2 expression in affected cells but also showed promise in reducing brain calcification and controlling phosphorus levels in animal models, highlighting a potential therapeutic approach for PFBC.

Background: Observational studies suggest that voluntary medical male circumcision (VMMC) may lower HIV risk among men who have sex with men (MSM). A randomized controlled trial (RCT) is needed to confirm this.

Objective: To assess the efficacy of VMMC in preventing incident HIV infection among MSM.

To improve the corrosion inhibition of zinc-rich epoxy (ZRE) composite coatings and shed light on the influence of the spatial structure of graphene fillers on the coatings' performance, three-dimensional graphene (3DG) and a conventional graphene sheet (G) were used to modify the ZRE composite paint, respectively. The effect of introducing the 2D G fillers on the anti-corrosion behavior of ZRE was studied comprehensively, and its optimal content was determined to be 0.5 wt%.

The 3C-like protease (3CLpro) is an essential enzyme for the replication of SARS-CoV-2 and other coronaviruses and thus is a target for coronavirus drug discovery. Nearly all inhibitors of coronavirus 3CLpro reported so far are covalent inhibitors. Here, we report the development of specific, noncovalent inhibitors of 3CLpro.

Background: Acupuncture has promising effects on chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS), but high-quality evidence is scarce.

Objective: To assess the long-term efficacy of acupuncture for CP/CPPS.

Design: Multicenter, randomized, sham-controlled trial.

Background: Postpartum hemorrhage is the most common cause of maternal death. Oxytocin is the standard therapy for the prevention of postpartum hemorrhage, but it requires cold storage, which is not available in many countries. In a large trial, we compared a novel formulation of heat-stable carbetocin with oxytocin.

Spontaneous symmetry breaking in quantum phase transitions leads to a system having degenerate ground states in its broken-symmetry phase. In order to detect all possible degenerate ground states for a broken-symmetry phase, we introduce a quantum fidelity defined as an overlap measurement between a system ground state and an arbitrary reference state. If a system has N-fold degenerate ground states in a broken-symmetry phase, the quantum fidelity is shown to have N different values with respect to an arbitrarily chosen reference state.
