A novel coumarin-based fluorescent probe LY was designed and synthesized in this work. LY could selectively recognize Cu via fluorescence quenching at 522 nm in a DMSO/HO solution. The recognition process experienced minimal interference from other common cations.

In this study, we induced progressive dopaminergic neuron loss in mice via rAAV-hSYN injection to cause the neuronal expression of α-synuclein, which produced neuroinflammation and behavioral alterations. We administered PLX5622, a colony-stimulating factor 1 receptor inhibitor, for 3 weeks prior to rAAV-hSYN injection, maintaining it for 8 weeks to eliminate microglia.

In this paper, a novel coumarin-derived fluorescent probe NY was designed and synthesized. NY displayed a significant ratiometric fluorescence response towards Cu in PBS buffer (10 mM, pH = 7.4), with the emission wavelength blue-shifted from 580 to 495 nm, and a fluorescence change from orange to green was evident under a 365 nm UV light.
