View Article and Find Full Text PDFAggregation of TAR-DNA binding protein-43 (TDP-43) is a pathological feature present in nearly 97 % cases of amyotrophic lateral sclerosis (ALS), making it an attractive target for pathogenic studies and drug screening. Here, we have performed a high-throughput screening of 1500 compounds from a natural product library and identified that lycorine, a naturally occurring alkaloid, significantly decreases the level of TDP-43 in a cellular model. We further demonstrate that lycorine reduces the level of TDP-43 both through inhibiting its synthesis and by promoting its degradation by the ubiquitin-proteasome system (UPS).

Porous membranes with uniform pore sizes have shown potential applications. Although the breath figure method is renowned for its simplicity, it requires high humidity conditions, presenting challenges in energy consumption and safety, especially in arid climates. This study introduces an approach that uses inorganic salt-based aerosols to successfully fabricate honeycomb-patterned porous membranes at low humidity levels, where pure water vapor cannot produce such structures.

Purkinje-related ventricular arrhythmias have been increasingly reported, and with the development of catheter ablation techniques, intervention for Purkinje-related arrhythmias has been shown to be effective. The characteristics of Purkinje fibres orientation in the 12 canine left ventricles were observed at a gross level by staining the endocardium with Lugol's solution. Purkinje fibres were observed microscopically by HE, Masson's, PAS glycogen, and Cx40 immunohistochemical staining.

In this study, an oxidative phosphorylation (OXPHOS) targeting nanoplatform was fabricated to alleviate hypoxia and enhance the performance of PDT by encapsulating IR780 and OXPHOS inhibitor atovaquone (ATO) in triphenylphosphine (TPP) modified poly(ethylene glycol) methyl ether-block-poly(L-lactide-co-glycolide) (mPEG-PLGA) nanocarriers (TNPs/IA). ATO by interrupting the electron transfer in OXPHOS could suppress mitochondrial respiration of tumor cells, economising on oxygen for the generation of ROS.
