Topical delivery of therapeutics on the skin can effectively alleviate skin symptoms of psoriasis and reduce systemic toxicity. However, the low delivery efficiency caused by the stratum corneum barrier limits the therapeutic impact. Here, we reported an oligopeptide hydrogel that encapsulates cell-penetrating-peptide (CPP)-decorated curcumin-loaded nanoemulsions (Cur-CNEs) to enhance the skin penetration of curcumin for topical treatment of psoriasis.

Background: Inositol-requiring enzyme 1 (IRE1) is an endoplasmic reticulum (ER)-resident transmembrane protein that senses ER stress and mediates an essential arm of the unfolded protein response (UPR). IRE1 reduces ER stress by upregulating the expression of multiple ER chaperones through activation of X-box-binding protein 1 (XBP1). Emerging lines of evidence have revealed that IRE1-XBP1 axis serves as a multipurpose signal transducer during oncogenic transformation and cancer development.

This paper delineates the structural features of AIM2 and its mechanisms of activation, emphasizing its capacity to detect cytosolic DNA and initiate inflammasome assembly. Additionally, we explore the diverse functions of AIM2 in different cells.
