Introduction: The aim was to evaluate the prognostic values of pretreatment 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography (PET) parameters for predicting local failure in nasopharyngeal carcinoma (NPC) patients in the intensity-modulated radiotherapy (IMRT) era.

Methods: Retrospective analysis was performed on 759 patients with NPC who underwent pretreatment 18F-FDG PET. The optimal cutoff values for maximum standardized uptake value (SUVmax) and metabolic tumor volume (MTV) were determined by receiver operating characteristic curve analysis.

Introduction: Temozolomide (TMZ) is a widely used chemotherapy agent for the treatment of malignant gliomas and other brain tumors. Despite its established therapeutic benefits, there is an ongoing need to understand better its safety profile, particularly in real-world clinical settings. This study aimed to identify critical adverse drug reactions (ADRs) associated with TMZ by utilizing the FDA Adverse Event Reporting System (FAERS) database, thereby providing valuable safety insights for clinical practice.

Antidepressants exhibit a considerable variation in efficacy, and increasing evidence suggests that individual genetics contribute to antidepressant treatment response. Here, we combined data on antidepressant non-response measured using rating scales for depressive symptoms, questionnaires of treatment effect, and data from electronic health records, to increase statistical power to detect genomic loci associated with non-response to antidepressants in a total sample of 135,471 individuals prescribed antidepressants (25,255 non-responders and 110,216 responders). We performed genome-wide association meta-analyses, genetic correlation analyses, leave-one-out polygenic prediction, and bioinformatics analyses for genetically informed drug prioritization.

: The aim of this study was to prepare monoclonal antibodies (mAbs) that broadly target and protect against infection by multi-drug-resistant (MDR) from different sources. : mAb 8E6 and mAb 1B5 were prepared by sequentially immunizing mice with a sublethal inoculation of three heterogeneous serotypes of pan-drug-resistant (PDR) , ST-208, ST-195, and ST-229. : The cross-recognition of heterogeneous bacteria (n = 13) by two mAbs and potential targets was verified, and the in vitro antibacterial efficacy of mAbs was assessed.

Article Synopsis
  • * The study focused on TDP-43, a protein associated with motor neuron diseases, using various experimental methods to explore how its phase behavior affects amyloid fibril formation.
  • * Findings suggest that TDP-43 can form aggregate intermediates that don't follow the traditional pathway to amyloid fibrils, indicating that the protein can cause disease regardless of the phase separation conditions.

Background: This study investigated the midterm primary patency of drug-coated balloons (DCBs), directional atherectomy plus balloon angioplasty (DA), and bare metal stent (BMS) angioplasty for the treatment of femoropopliteal lesions.

Methods: This two-center retrospective cohort study included 105 patients (110 limbs) undergoing DCB, DA, and BMS angioplasty-32 patients (34 limbs), 31 patients (32 limbs), and 42 patients (44 limbs), respectively. The demographic, baseline, and procedure data were collected, and the complications and midterm outcomes (patency, amputation-free survival, and clinically driven target lesion revascularization rates) were analyzed.

Proteins with intrinsically disordered regions (IDRs) often undergo phase separation to control their functions spatiotemporally. Changing the pH alters the protonation levels of charged sidechains, which in turn affects the attractive or repulsive force for phase separation. In a cell, the rupture of membrane-bound compartments, such as lysosomes, creates an abrupt change in pH.

Article Synopsis
  • The accumulation of CD8 T cells plays a significant role in liver injury and inflammation during metabolic dysfunction-associated steatohepatitis (MASH).
  • Empagliflozin (EMPA), a medication that inhibits SGLT2, showed effectiveness in reducing CD8 T cell levels and improving liver conditions in mice with MASH.
  • EMPA works by increasing β-hydroxybutyric acid, which inhibits factors that activate CD8 T cells, indicating that targeting these immune cells could be a promising approach for treating MASH in both mice and humans.

To evaluate the prognostic significance of the maximum standardized uptake value (SUVmax) in nasopharyngeal carcinoma (NPC), establish a gene signature that correlates with SUVmax, and explore the underlying biological behaviors associated with these correlations for the prediction of clinical outcomes. A cohort of 726 patients with NPC was examined to identify correlations between SUVmax and various clinical variables. RNA sequencing was performed to identify genes related to SUVmax, and these genes were used to develop an SUV signature.

Article Synopsis
  • Antidepressant effectiveness varies significantly among individuals, with genetics playing a role in how people respond to treatment.
  • A study analyzing data from 135,471 patients identified two new genetic locations (loci) linked to antidepressant non-response and confirmed the ability to predict treatment response using genetic information.
  • The research also explored potential new drug options that could help individuals who don't respond to traditional first-line antidepressants, highlighting the importance of larger sample studies for discovering relevant genetic factors.

Background: Diffusion kurtosis imaging (DKI) and neurite orientation dispersion and density imaging (NODDI) provide more comprehensive and informative perspective on microstructural alterations of cerebral white matter (WM) than single-shell diffusion tensor imaging (DTI), especially in the detection of crossing fiber. However, studies on systemic lupus erythematosus patients without neuropsychiatric symptoms (non-NPSLE patients) using multi-shell diffusion imaging remain scarce.

Methods: Totally 49 non-NPSLE patients and 41 age-, sex-, and education-matched healthy controls underwent multi-shell diffusion magnetic resonance imaging.

Article Synopsis
  • NAFLD is closely linked to type 2 diabetes (T2DM), and lifestyle changes are preferred treatments, but there's limited research on combining new glucose-lowering medications, semaglutide (a GLP-1 receptor agonist) and empagliflozin (an SGLT-2 inhibitor), for these patients.
  • This trial involves 105 overweight/obese participants and will last 52 weeks, comparing the effects of each drug alone and together on various health indicators, including liver health and blood sugar levels.
  • Findings from this study could shed light on the benefits of using both medications together in treating NAFLD associated with T2DM.

The purpose of this study was to establish an integrated predictive model that combines clinical features, DVH, radiomics, and dosiomics features to predict RIHT in patients receiving tomotherapy for nasopharyngeal carcinoma. Data from 219 patients with nasopharyngeal carcinoma were randomly divided into a training cohort (n = 175) and a test cohort (n = 44) in an 8:2 ratio. RIHT is defined as serum thyroid-stimulating hormone (TSH) greater than 5.

Three new sorbicillinoids sorbicatechols E-G (1-3), along with seven known compounds 4-10, were obtained from the ethanol extract of Penicillium sp. HS-11, a fungal endophyte of the medicinal plant Huperzia serrata. The structures of 1-3 were established by detailed interpretation of the spectroscopic data and their absolute configurations were established by comparative analyses of the ECD spectra.

The Taiwanese Government has long strived to accurately predict the short-term hourly concentration of PM for the warnings on air pollution. Long Short-Term Memory neural network (LSTM) based on deep learning improves the prediction accuracy of daily PM concentration but PM prediction for next hours still needs to be improved.
