Tumor organoids are important tools for cancer research, but current models have drawbacks that limit their applications for predicting response to therapy. Here, we developed a fast, efficient, and complex culture system (IPTO, individualized patient tumor organoid) that accurately recapitulates the cellular and molecular pathology of human brain tumors. Patient-derived tumor explants were cultured in induced pluripotent stem cell (iPSC)-derived cerebral organoids, thus enabling culture of a wide range of human tumors in the central nervous system (CNS), including adult, pediatric, and metastatic brain cancers.

A novel high-performance porous carbon material, lanthanum(iii)-doped camphor leaf-derived porous carbon (La/CPC), was synthesized and used as an adsorbent for the anionic dye Congo red (CR). The La/CPC composite was characterized by nitrogen adsorption and desorption isotherms, scanning electron microscopy, transmission electron microscopy and X-ray photoelectron spectroscopy. The adsorption performance of CR by the CPC and La/CPC composites with different contents of lanthanum(iii) was evaluated in fixed-bed breakthrough experiments and batch tests at room temperature (298 K).

The exploration of therapeutic targets in neuroblastoma (NB), which needs more attempts, can benefit patients with high-risk NB. Based on metabolomic and transcriptomic data in mediastinal NB tissues, we found that the content of long-chain acylcarnitine (LCAC) was increased and positively associated with leptin expression in advanced NB. Leptin over-expression forced naïve CD4+ T cells to differentiate into Treg cells instead of Th17 cells, which benefited from NB cell proliferation, migration, and drug resistance.

Background: Cutaneous melanoma (CM) is strongly associated with ultraviolet (UV) radiation, which contributes to the transformation of melanocytes into melanoma by inducing specific DNA damage. Here, we investigated the causal relationship between CM and genes related to sun-damaged skin, exploring specific target genes through various bioinformatics analyses.

Methods: The Gene Expression Omnibus (GEO) database was used to obtain differential genes for CM and normal skin, and the Genome-Wide Association Studies (GWAS) analysis offered summary-level melanoma data for CM.

Nanosecond pulsed electric fields (nsPEFs) can induce protein-mediated electroporation (PMEP) in voltage-gated ion channels. However, their effects on the tetrameric structure of voltage-gated potassium (Kv) channels remain unexplored. Our study pioneered the molecular dynamics (MD) investigation of the open-state (O) Kv channel to understand the effects of PMEP under unipolar and bipolar pulses (UP and BP).

The design of breeding programs is crucial for maximizing economic gains. Simulation provides the most efficient measures to test these programs, as real-world trials are often costly and time-consuming. We developed GOplan, a comprehensive and user-friendly R package designed to develop animal breeding programs considering pure-bred populations and crossbreeding systems.

Article Synopsis
  • - Chlorphenesin carbamate, a muscle relaxant, can metabolize into 4-chlorophenoxyacetic acid (4-CPA), which is banned by WADA, necessitating better identification methods in doping tests.
  • - A study analyzed urine samples from ten volunteers who ingested chlorphenesin carbamate, identifying a total of 29 metabolites through advanced mass spectrometry techniques.
  • - The findings indicate that two metabolites, M9 and M10, have longer detection windows, and the metabolic changes primarily involve histidine and β-alanine pathways, aiding in improving future doping control protocols.

Background: Acute-on-chronic liver failure (ACLF) is characterized by rapid onset, rapid development and a high short-term mortality rate. Systemic inflammation exerts an effect on the disease progression of ACLF.

Objectives: The purposes of this study were to explore the clinical significance that the inflammatory response has on the disease process of hepatitis B virus acute-on-chronic liver failure (HBV-ACLF) patients, to further compare the values of different inflammation-related biomarkers in the prognosis evaluation of HBV-ACLF patients, and to combine inflammatory-related markers to establish a new prediction model.

Article Synopsis
  • Pigs are a vital source of protein for humans, but the current reference genome limits our understanding of their genetic diversity due to structural variants (SVs).
  • This study created a comprehensive pangenome and genomic variation map from 599 deep-sequenced pig genomes across Eurasia, revealing significant differences among populations and identifying 546,137 SVs linked to distinct genomic features.
  • Notably, an SV in the TBX19 gene's promoter was identified, potentially influencing traits like growth and behavior in Asian pigs, highlighting the pangenome's utility for future genetic research.

Fifteen undescribed phenylpropanoyl phloroglucinol derivatives featuring modified isoprenyl and/or geranyl units (1-15), in conjunction with thirteen known analogues (16-28), were obtained from the glandular hairs of the fruit of Mallotus philippensis. Meanwhile, fourteen pairs of new enantiomers [(±)-1-9, (±)-11-15] were further resolved by chiral purification. Their structures were determined by comprehensive spectroscopic analyses, single-crystal X-ray diffraction experiments and quantum chemical calculations.

Biological variability poses significant challenges in the development of effective therapeutics, particularly when it comes to drug solubility and bioavailability. Poor solubility across varying physiological conditions often leads to reduced absorption and inconsistent therapeutic outcomes. This review examines how nanotechnology, especially through the use of nanomaterials and magnetic nanoparticles, offers innovative solutions to enhance drug solubility and bioavailability.

Unlabelled: AICAR (5-amino-4-imidazolecarboxyamide ribonucleoside), as a metabolic modulator, is classified in the S4 category by the World Anti-Doping Agency (WADA). Carbon Isotope Ratio Mass Spectrometry (CIR) is the mainstream method for distinguishing the endogenous and exogenous sources of AICAR in urine due to the significant individual difference in the concentration. The purpose of this study is to establish a gas chromatography combustion Isotope Ratio Mass Spectrometry (GC/C/IRMS) method for AICAR based on efficient two-dimensional liquid chromatography (2D-HPLC) separation.

: AICAR (5-amino-4-imidazolecarboxyamide ribonucleoside) was reported as the first pharmacological AMPK (adenosine 5'-monophosphate (AMP)-activated protein kinase) activator, and it has been confirmed to exhibit a significant endurance enhancement effect and prohibited for doping by the World Anti-Doping Agency. Due to the fact that the human body can produce such substances, in order to ensure fairness in sports competition, methods for rapid detection and multi-type identification of AICAR drugs taken orally should be established. : to assess AICAR levels, a new rapid, sensitive, efficient, and selective method was reported for the quantitative detection of AICAR in urine using LC-MS/MS.
