Although immune checkpoint blockade (ICB) therapies have shifted the treatment paradigm for non-small-cell lung cancer (NSCLC), many patients remain resistant. Here we characterize the tumor cell states and spatial cellular compositions of the NSCLC tumor microenvironment (TME) by analyzing single-cell transcriptomes of 232,080 cells and spatially resolved transcriptomes of tumors from 19 patients before and after ICB-chemotherapy. We find that tumor cells and secreted phosphoprotein 1-positive macrophages interact with collagen type XI alpha 1 chain-positive cancer-associated fibroblasts to stimulate the deposition and entanglement of collagen fibers at tumor boundaries, obstructing T cell infiltration and leading to poor prognosis.

The total synthesis of Kavaratamide A, a new Linear Lipodepsipeptide from the Marine Cyanobacterium was achieved by assembling the side chain in stage through condensation coupling, carbon-chain extension, Steglich esterification and Evans aldol reaction. In addition, total synthesis of Kavaratamide A's isomer, 5--kavaratamide A, is also successfully achieved. Cytotoxicity test suggested that Kavaratamide A and 5--Kavaratamide A have a moderate bioactivity against cancer cells.

Potassium (K) is the most abundant intracellular cation, but much remains unknown regarding how K homeostasis is integrated with other key bacterial biology aspects. Here, we show that K homeostasis disruption (CeoBC K uptake system deletion) impedes (Mtb) response to, and growth in, cholesterol, a critical carbon source during infection, with K augmenting activity of the Mtb ATPase MceG that is vital for bacterial cholesterol import. Reciprocally, cholesterol directly binds to CeoB, modulating its function, with a residue critical for this interaction identified.
