Assessing localization of the transient receptor potential vanilloid-1 (TRPV1) in skin nerve fibers is crucial for understanding its role in peripheral neuropathy and pain. However, information on the specificity and sensitivity of TRPV1 antibodies used for immunofluorescence (IF) on human skin is currently lacking. To find a reliable TRPV1 antibody and IF protocol, we explored antibody candidates from different manufacturers, used rat DRG sections and human skin samples for screening and human TRPV1-expressing HEK293 cells for further validation.

Article Synopsis
  • AL amyloidosis is a serious disease caused by the buildup of misfolded immune proteins, and birtamimab is an experimental antibody aimed at treating it by targeting toxic protein aggregates.
  • The VITAL clinical trial tested birtamimab in 260 newly diagnosed patients compared to a placebo, focusing on survival and hospitalization rates, but was halted early due to lack of significant overall benefits.
  • However, a post hoc analysis showed promising survival improvements for a subset of high-risk patients (Mayo stage IV), leading to further investigation in a new trial (AFFIRM-AL).

The effects of the long-term bilingual experience on structure and function of the cerebellum remain unclear. To explore whether there are differences in cerebellar gray matter structure between Cantonese-Mandarin bilinguals and Mandarin monolinguals and whether these different cerebellar structures have different resting-state functional connectivity (RSFC) with the cerebrum between the two groups, 30 Cantonese-Mandarin bilingual and 30 Mandarin monolingual college students were scanned by the T1-weighted magnetic resonance imaging (MRI) and resting-state functional MRI. Voxel-based morphology (VBM) analysis and RSFC analysis were used to analyze the cerebellar gray matter volume (GMV) and cerebellar-cerebro functional connectivity, respectively.

Cantonese and Mandarin are logographic languages, and the phonology is the main difference between the two languages. It is unclear whether the long-term experience of Cantonese-Mandarin bilingualism will shape different brain white matter structures related to phonological processing. A total of 30 Cantonese-Mandarin bilinguals and 30 Mandarin monolinguals completed diffusion-weighted imaging scan and phonological processing tasks.

Background: Many of genome features which could help unravel the often complex post-speciation evolution of closely related species are obscured because of their location in chromosomal regions difficult to accurately characterize using standard genome analysis methods, including centromeres and repeat regions.

Results: Here, we analyze the genome evolution and diversification of two recently diverged sister cotton species based on nanopore long-read sequence assemblies and Hi-C 3D genome data. Although D genomes are conserved in gene content, they have diversified in gene order, gene structure, gene family diversification, 3D chromatin structure, long-range regulation, and stress-related traits.

This paper examines the valuation effect of capital account liberalization. Using an event study approach and the policy announcement for RMB Qualified Foreign Institutional Investors (short for RQFII) as the event date, we find that overall, the stock market responded positively to the capital account liberalization announcement. In addition, we provide some heterogeneity that firms with more stringent financing constraints earn higher returns than their counterparts.

Background subtraction has attracted enormous interest in the field of moving object detection. However, when there are complex scenarios such as illumination changes, dynamic background, and noise, the moving object area obtained by background subtraction often has holes, noise, and shadows. This paper proposes a novel background update model based on matrix factorization, which uses the temporal continuity of video content to solve the problems of holes, noise, and shadows.

To develop more targeted intervention strategies, an important research goal is to identify markers predictive of clinical events. A crucial step toward this goal is to characterize the clinical performance of a marker for predicting different types of events. In this article, we present statistical methods for evaluating the performance of a prognostic marker in predicting multiple competing events.

In the past decade, several principal stratification-based statistical methods have been developed for testing and estimation of a treatment effect on an outcome measured after a postrandomization event. Two examples are the evaluation of the effect of a cancer treatment on quality of life in subjects who remain alive and the evaluation of the effect of an HIV vaccine on viral load in subjects who acquire HIV infection. However, in general the developed methods have not addressed the issue of missing outcome data, and hence their validity relies on a missing completely at random (MCAR) assumption.

Receiver operating characteristic (ROC) curves play a central role in the evaluation of biomarkers and tests for disease diagnosis. Predictors for event time outcomes can also be evaluated with ROC curves, but the time lag between marker measurement and event time must be acknowledged. We discuss different definitions of time-dependent ROC curves in the context of real applications.
