Article Synopsis
  • Histone H3 Lys36 (H3K36) methylation, particularly H3K36 dimethylation (H3K36me2) and trimethylation (H3K36me3), plays significant roles in DNA double-strand break (DSB) repair through two pathways: non-homologous end joining (NHEJ) and homologous recombination (HR).
  • Yeast cells lacking either H3K36me2 or H3K36me3 show reduced efficiency in DSB repair, with proteins yKu70 and Rfa1 interacting with these methylation forms to facilitate the repair process.
  • These findings indicate that different forms of H3K36 methylation are essential for

Background: As a chronic metabolic disease, diabetes poses a serious threat to human health and has become a major public health problem in China and worldwide. In 2020, 30% of Chinese people (aged ≥ 60 years) reported having diabetes mellitus. Moreover, individuals with diabetes living in rural areas face a significantly higher mortality risk compared to those in urban areas.

Background: Although percutaneous vertebral augmentation (PVA) is a commonly used procedure for treating vertebral compression fracture (VCF), the risk of vertebral refracture should be considered. Chronic kidney disease-mineral and bone disorder (CKD-MBD) is a systemic disease of mineral and bone metabolism. It is associated with an increased risk of fracture.

Stem cell therapy requires massive-scale homogeneous stem cells under strict qualification control. However, Prolonged expansion impairs the biological functions and results in senescence of mesenchymal stem cells (MSCs). We investigated the function of CTDSPL in the premature senescence process of MSCs and clarified that miR-18a-5p played a prominent role in preventing senescence of long-term cultured MSCs and promoting the self-renewal ability of MSCs.

Loss of transcription-coupled histone H3 lysine 36 trimethylation (H3K36me3) contributes to shorter lifespans in eukaryotes. However, the molecular mechanism of the decline of H3K36me3 during aging remains poorly understood. Here, we report that the degradation of the methyltransferase Set2 is the cause of decreased H3K36me3 levels during chronological aging in budding yeast.

Pyrano[4,3-]pyridine-diones, which are the key skeleton of bioactive compounds and functional materials, are usually prepared via a multistep synthesis using expensive substrates. This work demonstrates that Rh(III)-catalyzed dual C(sp)-H functionalization and C-O/C-N annulation of monoamide fumarates can produce pyrano[4,3-]pyridine-1,5(6)-diones in high yield (up to 82%) in a single step. The substrates of monoamide fumarates and acetylenes are structurally simple, readily available, and inexpensive.

Chimeric antigen receptor (CAR) T cell therapy is a novel therapeutic approach that uses gene editing techniques and lentiviral transduction to engineer T cells so that they can effectively kill tumors. However, CAR T cell therapy still has some drawbacks: many patients who received CAR T cell therapy and achieve remission, still had tumor relapse and treatment resistance, which may be due to tumor immune escape and CAR T cell dysfunction. To overcome tumor relapse, more researches are being done to optimize CAR T cell therapy to make it more precise and personalized, including screening for more specific tumor antigens, developing novel CAR T cells, and combinatorial treatment approaches.

Background: Systemic emphysematous infection caused by ( ) is a rare but severe infection which can be lethal if the diagnosis is delayed.

Case Summary: We report a rare case of systemic emphysematous infection hematogenous dissemination from a liver abscess caused by , complicated by multiple organ dysfunction syndrome, septic shock, bacteremia, emphysematous cystitis, prostate and left seminal vesicle abscesses in a diabetic patient. The patient simultaneously presented with spontaneous pneumoperitoneum secondary to rupture of the emphysematous liver abscess.

Aims: To explore the differentially expressed microRNAs (DEMs) in serum exosomes between gastric cancer (GC) patients and healthy people to provide new targets for GC diagnosis and treatment.

Methods: DEMs in serum exosomes were screened by microarray analysis and verified by RT-qPCR. The target genes of DEMs were predicted using Targetscan and miRTarBase databases and then overlapped with the DEGs of STAD in TCGA database to obtain the common target genes.

The concept of damage control surgery was first introduced in the 1990s. It has great significance in the treatment for critically ill patients, which not only greatly improves survival rate, but also helps doctors avoiding misdiagnosis and mistreatment. Herein, we present a case of gastric perforation caused by neoplasm with critical condition of the patient.

Methyltransferase Set2-mediated methylation of histone H3 lysine 36 (H3K36), which involves the addition of up to three methyl groups at this site, has been demonstrated to function in many chromatin-coupled events. The methylation of H3K36 is known to recruit different chromatin effector proteins, affecting transcription, mRNA splicing and DNA repair. In this study, we engineered two yeast set2 mutants that lack H3K36 mono/dimethylation (H3K36me1/2) and trimethylation (H3K36me3), respectively, and characterized their roles in the production of antisense transcripts under nutrient-rich conditions.

Circular ribonucleic acids (circRNAs) are widely expressed in human cells and play an important role in the pathogenesis of many diseases. Some circRNAs have microRNA (miRNA) binding response elements and interact with miRNA to regulate the expression of target genes.Four patients with a preliminary diagnosis of dengue fever (DF), peripheral whole blood sample in anticoagulant was collected before treatment (pretreatment group) and after effective treatment (posttreatment group), and eight samples were separated and used to screen differentially expressed circRNAs with microarray analysis.

  • Mitophagy is crucial for maintaining mitochondrial quality and cellular balance, and involves the Atg32 receptor system, but the regulatory pathways behind its expression are not well understood.
  • Bioinformatics research identifies the Paf1 complex (Paf1C) as a transcriptional repressor affecting the expression of certain genes, influencing mitophagy specifically under glucose starvation conditions.
  • Deletion of core Paf1C subunits enhances mitophagy activity, highlighting Paf1C's new role in modulating mitophagy at the transcriptional level in both yeast and mammals.

Background: Although promising results have recently been reported using dendritic cells (DCs) and cytokine-induced killer cells (CIKs) to treat pancreatic cancer (PC), its clinical effect and safety are associated with some controversy, and lack sufficient evidence. Here, we conducted a meta-analysis of 21 clinical trials to better evaluate the efficacy of DC-CIK immunotherapy in clinical practice to treat PC.

Methods: PubMed, Cochrane Library, China National Knowledge Infrastructure (CNKI) and Wanfang Data Knowledge Service Platform (WANFANG Data) were searched to identify clinical trials that used DC-CIK immunotherapy for PC.

Article Synopsis
  • MicroRNAs, particularly miR-122, are crucial in regulating gene expression and have been found to impact the effectiveness of cisplatin-based treatments for gastric cancer.
  • The study used various methods, including qRT-PCR and luciferase assays, to show that lower levels of miR-122 contribute to higher levels of excision repair cross-complementing 1 (ERCC1), leading to reduced treatment efficacy.
  • Overall, the findings suggest that diminished miR-122 expression promotes cisplatin resistance in gastric cancer by increasing ERCC1 expression, highlighting a potential target for improving treatment outcomes.

Background: Accumulating evidence indicated that long noncoding RNAs (lncRNAs) have a wide range of biological functions and may play significant roles in tumorigenesis and progression. However, the understanding of its functions and related competitive endogenous RNAs (ceRNAs) networks is much less than that of protein-coding genes, particularly in colon adenocarcinoma.

Methods: We comprehensively analyzed the sequencing data of protein-coding and noncoding RNAs in colon adenocarcinoma patients from The Cancer Genome Atlas (TCGA) database.
