Article Synopsis
  • Genomic alterations in tumors significantly influence cancer progression and treatment response, with whole-genome sequencing (WGS) emerging as a viable alternative to traditional targeted panel sequencing (TPS) due to advancements in cost and technology.
  • A study involving 120 cancer patients evaluated the clinical utility of WGS and found that 79% received genomic reports quickly, with 72% of these reports offering clinically relevant information.
  • The insights gained from WGS helped in selecting tailored treatments and clinical trials based on identifying specific mutations, potentially enhancing precision oncology and suggesting routine integration of WGS in cancer management.

Article Synopsis
  • The study examines changes in North Pacific water masses over the last 60 years, emphasizing the competition between anthropogenic aerosols (AAs) and greenhouse gases (GHGs).
  • During the period from 1950 to 1985, AAs primarily influenced salinity changes, but after 1985, GHGs became the dominant factor.
  • The research highlights the connection between surface changes driven by these climate forces and deeper ocean patterns, illustrating the significant impact of AAs and GHGs on ocean thermocline dynamics.

Atlantic Meridional Overturning Circulation (AMOC) exhibits interdecadal to multidecadal variability, yet the role of surface freshwater flux (FWF) variability in this AMOC variability remains unclear. This study isolates the contribution of FWF variability in modulating AMOC through a partially coupled experiment, in which the effect of the interactive FWF is disabled. It is demonstrated that the impact of the coupled FWF variability enhances the persistence of density and deep convection anomalies in the Labrador Sea (LS), thus lengthening the period of the AMOC oscillation on multidecadal timescale and suppressing its ∼30-year periodicity.

Cellular dynamics and fate decision in early human embryogenesis remain largely unknown owing to the challenges of performing studies in human embryos. Here, we explored whole-genomes of 334 single-cell colonies and targeted deep sequences of 379 bulk tissues obtained from various anatomical locations of seven recently deceased adult human donors. Using somatic mutations as an intrinsic barcode, we reconstructed early cellular phylogenies that demonstrate (1) an endogenous mutational rate that is higher in the first cell division but decreases to approximately one per cell per cell division later in life; (2) universal unequal contribution of early cells to embryo proper, resulting from early cellular bottlenecks that stochastically set aside epiblast cells within the embryo; (3) examples of varying degrees of early clonal imbalances between tissues on the left and right sides of the body, different germ layers and specific anatomical parts and organs; (4) emergence of a few ancestral cells that will substantially contribute to adult cell pools in blood and liver; and (5) presence of mitochondrial DNA heteroplasmy in the fertilized egg.

A coordinated set of large ensemble atmosphere-only simulations is used to investigate the impacts of observed Arctic sea ice-driven variability (SIDV) on the atmospheric circulation during 1979-2014. The experimental protocol permits separating Arctic SIDV from internal variability and variability driven by other forcings including sea surface temperature and greenhouse gases. The geographic pattern of SIDV is consistent across seven participating models, but its magnitude strongly depends on ensemble size.

Somatic genome mutations occur due to combinations of various intrinsic/extrinsic mutational processes and DNA repair mechanisms. Different molecular processes frequently generate different signatures of somatic mutations in their own favored contexts. As a result, the regional somatic mutation rate is dependent on the local DNA sequence, the DNA replication/RNA transcription dynamics and epigenomic chromatin organization landscape in the genome.

Recent studies documenting shifts in spatial distribution of many organisms in response to a warming climate highlight the need to understand the mechanisms underlying species distribution at large spatial scales. Here we present one noteworthy example of remote oceanographic processes governing the spatial distribution of adult silver hake, Merluccius bilinearis, a commercially important fish in the Northeast US shelf region. Changes in spatial distribution of silver hake over the last 40 years are highly correlated with the position of the Gulf Stream.

Histologic and clinicopathologic findings of a woodchuck (Marmota monax) vertically infected with woodchuck hepatitis virus (WHV) are presented. The liver exhibits marked cirrhotic changes, which is characteristic of the pre-transformation phase of WHV. At necropsy, the woodchuck exhibited ascites and the liver had a grossly nodular appearance.
