Neuroimaging genetics is a powerful approach to jointly explore genetic features with rich brain imaging phenotypes for neurodegenerative diseases. Conventional imaging genetics approaches based on canonical correlation analysis cannot accommodate multimodal inputs effectively and have limited interpretability. We propose a novel imaging genetics approach based on non-negative matrix factorization (NMF).

Aims: It remains unclear if patients with acute heart failure syndrome (AHFS) need to reach the maximally tolerated doses of renin-angiotensin system blockers (RASBs) or beta-blockers (BBs) to obtain a survival benefit. This study evaluated the dose-response relationship between RASBs or BBs and survival in AHFS patients.

Methods And Results: In total, 5331 patients in the Korean Acute Heart Failure registry were analysed based on the doses of RASBs and BBs at discharge.

Extracellular potassium (K) homeostasis is achieved by a concerted effort of multiple organs and tissues. A limitation in studies of K homeostasis is inadequate techniques to quantify K fluxes into and out of organs and tissues in vivo. The goal of the present study was to test the feasibility of a novel approach to estimate K distribution and fluxes in vivo using stable K isotopes.

An extracellular matrix (ECM) membrane made up of ECM hydrogels has great potentials to develop a physiologically relevant organ-on-a-chip because of its biochemical and biophysical similarity tobasement membranes (BMs). However, the limited mechanical stability of the ECM hydrogels makes it difficult to utilize the ECM membrane in long-term and dynamic cell/tissue cultures. This study proposes a thin but robust and transparent ECM membrane reinforced with silk fibroin (SF)/polycaprolactone (PCL) nanofibers, which is achieved byself-assembly throughout a freestanding SF/PCL nanofiber scaffold.

Article Synopsis
  • A multicenter study examined the long-term effects of bilateral subthalamic deep brain stimulation (DBS) on Parkinson's disease patients with SNCA mutations, highlighting that data on this topic is limited.
  • Out of four patients studied, three with SNCA duplication had positive outcomes after an average of 5.4 years post-DBS, while one with a different mutation experienced deterioration in cognitive and psychiatric functions despite some motor benefits.
  • The results suggest that while DBS can help with motor fluctuations in certain PD patients, cognitive and psychiatric health significantly impacts long-term results.

Background: Sudden cardiac death (SCD) and stroke-related events accompanied by atrial fibrillation (AF) can affect morbidity and mortality in hypertrophic cardiomyopathy (HCM). This study sought to evaluate a scoring system predicting cardio-cerebral events in HCM patients using cardiopulmonary exercise testing (CPET).

Methods: We investigated the role of a previous prediction model based on CPET, the HYPertrophic Exercise-derived Risk score for Heart Failure-related events (HyperHF), which is derived from peak circulatory power ventilatory efficiency and left atrial diameter (LAD), for predicting a composite of SCD-related (SCD, serious ventricular arrhythmia, death from cardiac cause, heart failure admission) and stroke-related (new-onset AF, acute stroke) events.

Article Synopsis
  • Pediatric hemato-oncologic patients often need central catheters for chemotherapy, with the cavoatrial junction being the preferred insertion site, but existing identification methods lack standardization.
  • A study involving 15 oncology patients utilized augmented reality to create 3D models for precise catheter placement and confirmed the positioning with portable x-rays.
  • Results showed that the augmented reality method significantly reduced the average distance from the cavoatrial junction compared to traditional techniques, indicating it is a safer and more accurate approach without extra costs.

B-cell-depleting therapies may lead to prolonged disease and viral shedding in individuals infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and this viral persistence raises concern for viral evolution. We report sequencing of early and late samples from a 335-day infection in an immunocompromised patient. The virus accumulated a unique deletion in the amino-terminal domain of the spike protein, and complete deletion of ORF7b and ORF8, the first report of its kind in an immunocompromised patient.
