We have identified two distinct Pax8 (a and b) mRNAs from the thyroid gland of the rainbow trout (Oncorhynchus mykiss), which seemed to be generated by alternative splicing. Both Pax8a and Pax8b proteins were predicted to possess the paired domain, octapeptide, and partial homeodomain, while Pax8b lacked the carboxy-terminal portion due to an insertion in the coding region of the mRNA. RT-PCR analysis showed each of Pax8a and Pax8b mRNAs to be abundantly expressed in the thyroid and kidney.

FoxF1 is a member of the winged helix/forkhead transcription factor gene family. We have cloned the cDNA encoding a rainbow trout FoxF1 homologue, and examined its developmental gene expression pattern. By 7 days postfertilization (dpf at 14 degrees C), FoxF1 is expressed throughout the alimentary tract in the mesenchymal cells adjacent to the endodermal epithelium, with intense signals on the dorsal side of the oral cavity and in the primitive stomach.

We have cloned two distinct cDNAs encoding salmon-type calcitonin (sCT)-I cDNAs from the ultimobranchial gland of rainbow trout, Oncorhynchus mykiss. Both cDNAs were predicted to encode nearly identical sCT-I precursors which consisted of an N-terminal peptide of 80 amino acid residues, a putative cleavage site Lys-Arg, sCT-I, a cleavage and amidation sequence Gly-Lys-Lys-Arg, and a C-terminal peptide of 18 amino acids. Development of sCT-I-expressing cells was then examined by employing conventional histochemical staining, in situ hybridization with a specific cRNA probe, and further immunohistochemistry.

Four types of calcitonin are produced in salmonid fish, although their functional diversity is almost unknown. To explore the significance of these isoforms, we have characterized salmon-type calcitonin (sCT) mRNAs in the rainbow trout (Oncorhynchus mykiss), and examined their tissue distribution. In addition to the previously isolated sCT-I cDNAs, two new forms of sCT cDNA were cloned from the ultimobranchial gland, and one of them (sCT-IV cDNA) was predicted to encode an N-terminal peptide of 80 amino acid residues, a putative cleavage site Lys-Arg, sCT-IV, a cleavage and amidation sequence Gly-Lys-Lys-Arg, and a C-terminal peptide of 18 amino acids.

In amphibians, calcium carbonate crystals are present in the endolymphatic sac and the inner ear. The formation of these crystals is considered to be facilitated by a protein called otoconin-22. Little is known about the influence of hemodynamic changes on ventricular performance during pregnancy in patients with congenital heart disease. A prospective evaluation was made of 17 pregnancies in 11 women with repaired tetralogy of Fallot (ToF). Twenty age-matched non-pregnant women and 41 stable pregnant women without heart disease were studied as controls.
