Since trastuzumab was approved in 2012 for the first-line treatment of gastric cancer (GC), no significant advancement in GC targeted therapies has occurred. Synthetic lethality refers to the concept that simultaneous dysfunction of a pair of genes results in a lethal effect on cells, while the loss of an individual gene does not cause this effect. Through exploiting synthetic lethality, novel targeted therapies can be developed for the individualized treatment of GC.

Colorectal cancer (CRC) remains a primary cause of cancer mortality globally, necessitating precise prognostic indicators for effective clinical management. Our study introduces the Senescence Risk Score (SRRS), based on several senescence-related genes (SRGs), a potent prognostic tool designed to measure cellular senescence in CRC. The higher SRRS predicts a poorer prognosis, providing a novel and efficient approach to patient stratification.

Treatment of the 2,5-bis(2'-pyridyl)pyrrolato (PDP) anion with {Ru(COD)Cl} in THF readily yielded [Ru(PDP)(COD)Cl] (1) in almost quantitative yield. Anion metathesis of 1 in organic solvent by NO and OTf (OTf = triflato) gave [Ru(PDP)(COD)(NO)] (2) and [Ru(PDP)(COD)(OTf)] (3), and in aqueous solution by BF and PF afforded aqueous complexes [Ru(PDP)(COD)(HO)](BF) (4+·BF) and [Ru(PDP)(COD)(HO)](PF) (4+·PF), respectively. Treatment of 1 with PhICl in CHCl afforded 5 with halogenated pyrrole.
