Article Synopsis
  • The study aimed to assess the effectiveness of combining PD-1 inhibitors with chemotherapy in patients with recurrent or metastatic nasopharyngeal carcinoma (R/M NPC).
  • A total of 142 patients were analyzed, showing that the combination treatment led to significantly higher response and remission rates compared to chemotherapy alone, though it also resulted in increased instances of certain side effects.
  • Key factors like treatment type, age, and treatment duration were identified as important in predicting patient survival, with a model developed that accurately forecasted progression-free survival for 1 and 2 years.

The selection of scaffold materials and the optimization of scaffold morphological and mechanical properties are critical for successful bone tissue engineering. We fabricated porous scaffolds of nano-sized zirconia using a replication technique. The study aimed to explore the relationship between porosity, pore size, mechanical strength, cell adhesion, and cell proliferation in the zirconia scaffolds.

Background: Notch-1 promotes invasion and metastasis of cancer cells but its role in salivary adenoid cystic carcinoma (SACC) remains unelucidated. Here, we sought to investigate the effect of Notch-1 knockdown on the invasion and metastasis of SACC cells.

Methods: Stable ACC-M cells whose Notch-1 was silenced by lentiviral vectors were established.

Objective: To construct and identify a lentiviral vector of RNA interference targeting human Notch-1 gene.

Methods: To determine the Notch-1 gene sequences, three RNAi target sequences (shRNA1-3) were designed in accordance with the RNAi sequence design principles and cloned into the lentiviral vector pLenOR-THM by endonuclease BamH I restriction, EcoR I double digestion, and T4 DNA-ligase ligation. After the transformation into competent DH5alpha bacteria, the candidate clones were identified by Kpn I and EcoR I double digestion and DNA sequencing.
