Article Synopsis
  • Arginase contributes to the development of an immunosuppressive environment in colorectal cancer, making the disease less responsive to immunotherapy; its activity in macrophages skews them toward a less effective M2-like polarization.
  • The study introduced a microparticle formulation (PG/ICG@MPs) that combines the natural compound piceatannol 3'-O-glucoside and indocyanine green to target tumors and modify immune responses through inhibiting arginase and enhancing nitric oxide levels.
  • Results showed that PG/ICG@MPs effectively promote the repolarization of macrophages towards the M1 phenotype and boost tumor immunity, demonstrating promising potential for combined cancer therapies that can be translated into clinical

Background: Colorectal cancer (CRC) patients with mismatch repair-deficient/microsatellite instability-high (dMMR/MSI-H) status are conventionally perceived as unresponsive to adjuvant chemotherapy (ACT). The mitochondrial transcription factor A (TFAM) is required for mitochondrial DNA copy number (mtDNA-CN) expression. In light of previous findings indicating that the frequent truncating-mutation of TFAM affects the chemotherapy resistance of MSI CRC cells, this study aimed to explore the potential of mtDNA-CN as a predictive biomarker for ACT efficacy in dMMR CRC patients.

Circular RNAs are a class of covalently closed single-stranded loop RNAs that have been implicated to play a functional role in almost all types of cancers. Previous studies have revealed that circMYBL2 acts as a tumor-promoting circular RNA. In this study, we found that circMYBL2 in colorectal cancer encodes a 185-amino acid protein, p185.

Untethered capsules hold clinical potential for the diagnosis and treatment of gastrointestinal diseases. Although considerable progress has been achieved recently in this field, the constraints imposed by the narrow spatial structure of the capsule and complex gastrointestinal tract environment cause many open-ended problems, such as poor active motion and limited medical functions. In this work, we describe the development of small-scale magnetically driven capsules with a distinct magnetic soft valve made of dual-layer ferromagnetic soft composite films.

Despite the notable achievements of programmed death 1 (PD-1) antibodies in treating various cancers, the overall efficacy remains limited in the majority of colorectal cancer (CRC) cases. Metabolism reprogramming of tumors inhibits the tricarboxylic acid (TCA) cycle, leading to down-regulation of fumarate hydratase (FH), which is related to poor prognosis in CRC patients. By establishing a tumor-bearing mouse model of CRC with Fh1 expression deficiency, we confirmed that the therapeutic effect of PD-1 antibodies alone was suboptimal in mice with low Fh1 expression, which was improved by combination with a protein invertase subtilisin/kexin 9 (PCSK9) inhibitor.

Article Synopsis
  • This study investigates how body composition, specifically skeletal muscle and fat levels measured via CT scans, impacts postoperative complications and long-term outcomes in rectal cancer patients after surgery.
  • Researchers analyzed data from 415 patients at Wuhan Union Hospital between 2014 and 2018, using CT images taken within 3 months before surgery to assess muscle and fat indices.
  • Findings show that higher skeletal muscle density and index correlate with better overall survival and disease-free survival, while larger intermuscular fat areas are linked to poorer outcomes, suggesting body composition is crucial for predicting recovery and prognosis in these patients.

Article Synopsis
  • This study explored the connection between gut microbiota changes and colorectal cancer (CRC) in patients with intestinal obstruction.
  • The research involved 313 CRC patients, revealing that those with obstruction had shorter survival rates and higher microbial diversity in tumor tissues compared to those without obstruction.
  • Specifically, the Bacteroidetes phylum was notably more abundant in the tumors of patients with obstruction, suggesting a potential link between this microbiota shift and poorer prognosis in CRC.

Background: The tumor microenvironment (TME) is an important factor that regulates the progression of colorectal cancer (CRC). Cancer-associated fibroblasts (CAFs) are the main mesenchymal cells in the TME and play a vital role in tumor progression; however, the specific underlying mechanisms require further study.

Methods: Multiple single-cell and transcriptome data were analyzed and validated.

Background: Mesenchymal stromal cells (MSCs) have attracted great attention in the application of cell-based therapy because of their pluripotent differentiation and immunomodulatory ability. Due to the limited number of MSCs isolated from donor tissues, a large number of MSCs need to be expanded in a traditional two-dimensional cell culture device to obtain a sufficient therapeutic amount. However, long-term cultivation of MSCs in vitro has been proven to reduce their differentiation potential and change their immunomodulatory characteristics.
