Introduction: Relapsed or refractory extranodal natural killer/T-cell lymphoma (R/R ENKTL) is a rare and aggressive subtype arising from natural killer or cytotoxic T-cells, predominantly affecting the nasal cavity and paranasal sinuses, lacking a standardized therapeutic approach. Sugemalimab, a fully human, full-length anti-PD-L1 immunoglobulin G4 (IgG4) monoclonal antibody (mAb), has been investigated in a Single-Arm, Multicenter, Phase II Study (GEMSTONE-201). The results demonstrated significant efficacy, favorable tolerability, and manageable adverse reactions of sugemalimab in R/R ENKTL.

Article Synopsis
  • - The study investigates how serum magnesium levels affect the effectiveness and survival outcomes of immune checkpoint blockers (ICBs) in cancer therapy, highlighting the need for more research on this relationship.
  • - In a multi-center analysis of 1,441 cancer patients treated with ICBs, it was found that higher serum magnesium levels (≥ 0.79 mmol/L) correlated with better responses and longer progression-free and overall survival compared to lower levels.
  • - The findings suggest that serum magnesium levels are an independent prognostic factor in cancer patients undergoing ICB therapy, but more prospective studies are necessary to validate these results.

Recurrent abnormalities in immune surveillance-related genes affect the progression of diffuse large B-cell lymphoma (DLBCL) and modulate the response to therapeutic interventions. CD58 interacts with the CD2 receptor on T cells and NK cells and is recurrently mutated and deleted in DLBCL, suggesting that it may play a role in regulating antitumor immunity. In this study, we comprehensively analyzed the genomic characteristics of CD58 through targeted next-generation sequencing, RNA sequencing (RNA-seq), whole-exome sequencing, and single-cell RNA-seq in patients with newly diagnosed DLBCL.
