This study intents to detect graphical network features associated with seizure relapse following antiseizure medication (ASM) withdrawal. Twenty-four patients remaining seizure-free (SF-group) and 22 experiencing seizure relapse (SR-group) following ASM withdrawal as well as 46 matched healthy participants (Control) were included. Individualized morphological similarity network was constructed using T1-weighted images, and graphic metrics were compared between groups.

Recurrent missense mutations in the human epidermal growth factor receptor 2 (HER2) have been identified across various human cancers. Among these mutations, the active S310F mutation in the HER2 extracellular domain stands out as not only oncogenic but also confers resistance to pertuzumab, an antibody drug widely used in clinical cancer therapy, by impeding its binding. In this study, we have successfully employed computational-aided rational design to undertake directed evolution of pertuzumab, resulting in the creation of an evolved pertuzumab variant named Ptz-SA.

Group B (GBS) is a leading pathogen responsible for fatal infections in newborns primarily due to vertical transmission from colonized mothers. Cases of invasive GBS infections in adults have also increased and attracted attention recently. To comprehensively understand the evolving burden of vaginal GBS carriage in pregnant and non-pregnant women, as well as the trends in invasive GBS diseases and antibiotic resistance in China, we conducted a retrospective study using data from a large tertiary hospital in Beijing from 2013 to 2023.

Yellow tepal is a unique trait of the American lotus (Nelumbo lutea), and all yellow lotus cultivars in the market possess genetic material from the American lotus. However, the formation of yellow tepals in lotus and the genetic mechanism of their formation remain unclear. In this study, we identified a transposon DNA/hAT-Ac, located within the promoter region of an R2R3-MYB transcription factor, MYB12, by comparing the insertion patterns of transposons in the genomes of American and Asian lotus (Nelumbo nucifera).

Article Synopsis
  • The study focuses on Ethylene-responsive factor (ERF) transcription factors in Nelumbo nucifera, an important aquatic plant, highlighting their crucial roles in plant development and stress responses.
  • Researchers identified 107 ERF genes in the N. nucifera genome, categorized into 11 groups with varying expression patterns across different tissues; some genes show responsiveness to abiotic stresses like cold, drought, and salinity.
  • The findings offer significant insights for future research on the functional roles of NnERF genes, especially in enhancing stress tolerance in N. nucifera.

  • The text discusses a group of emerging human fungal pathogens that are resistant to multiple drugs and have been increasingly isolated in countries like China, Malaysia, and Brazil.
  • These fungi utilize various strategies, such as biofilm formation and immune evasion, to infect hosts and cause disease, along with a concerning resistance to antifungal treatments like amphotericin B (AMB).
  • The review aims to enhance our understanding of these fungi's virulence factors, how AMB works against them, and why they are resistant to this common antifungal medication.

Thalassemia is a hemoglobin disease characterized by reduced or complete absence of the production of the α/β globin gene. Currently, the detection of β-thalassemia carriers is based on differences in blood cell parameters. However, β-thalassemia carriers cannot be distinguished from α- and β-thalassemia co-inherited carriers based solely on hematological findings, and the differential diagnosis must rely on molecular diagnosis.

  • A multicenter study across 65 hospitals in China analyzed the effectiveness of contezolid against various strains of Staphylococcus and Enterococcus, focusing on its susceptibility and resistance patterns.
  • The research found that all strains were susceptible to contezolid, with low resistance rates to other antibiotics, notably finding higher linezolid resistance in Enterococcus faecalis than in Enterococcus faecium.
  • Results showed contezolid had a lower MIC50 compared to linezolid, indicating it may be a more effective option against resistant bacterial strains, particularly methicillin-resistant Staphylococcus and linezolid-resistant E. faecalis.

  • Carbapenem-resistant bacteria (CRB), including strains like CRAB, CRPA, and CRE, are a major public health concern in China, with the aim of this study being to evaluate their prevalence and antimicrobial susceptibility.
  • Researchers collected 7,194 strains of gram-negative bacteria and identified 924 carbapenem-resistant strains, testing their susceptibility to 21 antimicrobial agents using broth microdilution and analyzing carbapenemase genes.
  • Results showed varying isolation rates for CRE, CRAB, and CRPA, with aztreonam/avibactam, colistin, and eravacycline proving highly effective against them, particularly outperforming ceftazidime/avibactam for certain strains. *

Strains from the species complex (CGSC) have caused the Pacific Northwest cryptococcosis outbreak, the largest cluster of life-threatening fungal infections in otherwise healthy human hosts known to date. In this study, we utilized a pan-phenome-based method to assess the fitness outcomes of CGSC strains under 31 stress conditions, providing a comprehensive overview of 2,821 phenotype-strain associations within this pathogenic clade. Phenotypic clustering analysis revealed a strong correlation between distinct types of stress phenotypes in a subset of CGSC strains, suggesting that shared determinants coordinate their adaptations to various stresses.

  • The study presents the first detailed genome analysis of a rare human yeast pathogen called Candida mucifera, identified in a patient suffering from chronic ear infections.
  • Researchers found that this pathogen shows lower effectiveness to azole antifungals, which are commonly used to treat fungal infections.
  • The findings suggest that Candida mucifera has similarities in drug resistance to its related species in the Trichomonascus ciferrii complex.

Inadequate antigen-specific T cells activation hampers immunotherapy due to complex antigen presentation. In addition, therapeutic T cell expansion is constrained by slow expansion rates and limited functionality. Herein, we introduce a model fusion protein termed antigen-presenting cell-mimic fusion protein (APC-mimic), designed to greatly mimicking the natural antigen presentation pattern of antigen-presenting cells and directly expand T cells both and .
