Photodynamic therapy (PDT) using traditional type II photosensitizers (PSs) has been limited in hypoxic tumors due to excessive oxygen consumption. The conversion from type II into a less oxygen-dependent type I PDT pathway has shown the potential to combat hypoxic tumors. Herein, the design of a heterodimeric PS, NBSSe, by conjugating a widely used type I PS NBS and a type II PS NBSe via molecular dimerization, achieving the aggregation-regulated efficient type I photodynamic conversion for the first time is reported.

Biofilm-induced chronic bacterial infections represent a significant challenge in modern medicine due to their resistance to conventional antibiotic treatments. Cell membrane genetic engineering has been utilized to confer cell membranes with functionalities for diagnostic and therapeutic purposes but concerns over cost and variable modification results. Fluorescence-image guided surgery (FGS) can intraoperatively provide real-time visualization of a tumor incisal edge and high-resolution identification of tumor foci to improve treatment outcomes. In this contribution, we report a fluorescent probe NB-TAM based on intramolecularly folded photoinduced electron transfer (PET), which displayed a prominent turn-on response in the near-infrared (NIR) window upon specific interaction with the estrogen receptor (ER). Significantly, NB-TAM could delineate a clear tumor incisal edge (tumor-to-normal tissue ratio > 5) in a 70-min time window, and was successfully used to guide the facile and precise resection of ER+ breast tumors in mice.
