R-loops are critical structures that play pivotal roles in regulating genomic stability and modulating gene expression. This study investigates the interactions between the 5-methylcytosine (mC) methyltransferase NOP2/Sun RNA methyltransferase 2 (NSUN2) and R-loops in the transcriptional dynamics and damage repair process of bladder cancer (BCa) cells. We observed markedly elevated levels of R-loops in BCa cells relative to normal urothelial cells.

Prostate cancer (PCa) gradually becomes the most common cancer in men in many countries, of which circRNAs and methylated modification exert an essential role in PCa progression. However, the concrete mechanisms of N6-methyladenosine (m6A) modification of circRNAs in PCa remain unclear. In our study, we identified circRPS6KC1, a novel and up-regulated circular RNA in PCa, through circRNA sequencing.

Article Synopsis
  • Chronic infection with Hepatitis B Virus (HBV) leads to weak virus-specific T cell responses, complicating viral clearance while causing significant liver inflammation due to immune responses.
  • A new assay developed for detecting HBV Capsid-Antibody Complexes (CACs) showed high accuracy in measuring these complexes in serum and linked them to increased liver inflammation and damage in chronic hepatitis B patients.
  • The study provides strong evidence that CACs play a key role in complement-mediated liver injury, establishing a new factor to consider alongside existing clinical markers for monitoring inflammation in chronic hepatitis B.

Iron and 2-oxoglutarate dependent (Fe/2OG) enzymes exhibit an exceedingly broad reaction repertoire. The most prevalent reactivity is hydroxylation, but many other reactivities have also been discovered in recent years, including halogenation, desaturation, epoxidation, endoperoxidation, epimerization, and cyclization. To fully explore the reaction mechanisms that support such a diverse reactivities in Fe/2OG enzyme, it is necessary to utilize a multi-faceted research methodology, consisting of molecular probe design and synthesis, in vitro enzyme assay development, enzyme kinetics, spectroscopy, protein crystallography, and theoretical calculations.

  • Probiotics, particularly from yogurt, may help improve sleep quality and reduce disturbances among adults, as suggested by a study analyzing data from over 49,000 participants in the National Health and Nutrition Examination Survey (NHANES).
  • About 14% of the participants consumed yogurt or probiotic supplements, with findings indicating that this group had a lower prevalence of sleep disorders compared to those who did not consume these foods.
  • The study highlights significant associations between probiotic intake and better sleep outcomes, especially in certain demographics such as males and individuals with normal BMI, indicating that including yogurt in the diet could be beneficial for sleep health.

Single gamete cell sequencing together with long-read sequencing can reliably produce chromosome-level phased genomes. In this study, we employed PacBio HiFi and Hi-C sequencing on a male Landrace pig, coupled with single-sperm sequencing of its 102 sperm cells. A haplotype assembly method was developed based on long-read sequencing and sperm-phased markers.

Background: As a highly heterogeneous tumor, non-small cell lung cancer (NSCLC) is famous for its high incidence and mortality worldwide. Smoking can cause genetic changes, which leading to the occurrence and progress of NSCLC. Nevertheless, the function of smoking-related genes in NSCLC needs more research.

R-loops are prevalent three-stranded nucleic acid structures, comprising a DNA-RNA hybrid and a displaced single-stranded DNA, that frequently form during transcription and may be attributed to genomic stability and gene expression regulation. It was recently discovered that RNA modification contributes to maintain the stability of R-loops such as N6-methyladenosine (mA). Yet, mA-modified R-loops in regulating gene transcription remains poorly understood.

Objective: To examine the correlation of neutrophil CD64 (nCD64) index with neurosyphilis (NS) across different stages of syphilis.

Methods: A total of 1243 syphilis patients at different stages (344 of primary, 385 of secondary, and 514 of tertiary) included in this study were divided into NS and non-NS (NNS). Correlations of nCD64 index with currently used syphilis biomarkers were explored using Spearman correlation test.

Several studies have indicated that circular RNAs (circRNAs) play vital roles in the progression of various diseases, including bladder cancer (BCa). However, the underlying mechanisms by which circRNAs drive BCa malignancy remain unclear. In this study, we identified a novel circRNA, circPSMA7 (circbaseID:has_circ_0003456), showing increased expression in BCa cell lines and tissues, by integrating the reported information with circRNA-seq and qRT-PCR.

Fluorescent lateral flow immunoassay (LFA) systems are versatile tools for sensitive and quantitative detection of disease markers at the point of care. However, traditional fluorescent nanoparticle-based lateral flow immunoassays are not visible under room light, necessitate an additional fluorescent reader, and lack flexibility for different application scenarios. Herein, we report a dual-readout LFA system for the rapid and sensitive detection of C-reactive protein (CRP) in clinical samples.

This article offers an optimal control tracking method using an event-triggered technique and the internal reinforcement Q-learning (IrQL) algorithm to address the tracking control issue of unknown nonlinear systems with multiple agents (MASs). Relying on the internal reinforcement reward (IRR) formula, a Q-learning function is calculated, and then the iteration IRQL method is developed. In contrast to mechanisms triggered by time, an event-triggered algorithm reduces the rate of transmission and computational load, since the controller may only be upgraded when the predetermined triggering circumstances are met.

Over 240 million people worldwide are chronically infected with Hepatitis B Virus (HBV), a hepatotropic DNA virus with an evolutionary root of over 400 million years. Persistent HBV infection exhibits distinct and diverse phases of disease, from minimal liver pathology to fulminant Hepatitis, that vary in duration and severity among individuals. Although huge progress has been made in HBV research which has yielded an effective prophylactic vaccine and potent antiviral therapy, our understanding of its virology and immunobiology is still far from complete.
