Cell-permeable ubiquitin (Ub) probes labeled with a fluorescent or biotin tag have become effective tools for studying the activity of deubiquitinating enzymes (DUBs) in living cells. However, such probes are usually obtained through total chemical synthesis. In this study, we develop a Sortase A (SrtA)-mediated semi-synthetic strategy for preparing cell-permeable Ub-propargylamide (Ub-PA) with a fluorescent or biotin tag.

Palladium/Norbornene Cooperative Catalysis 2-Fold C-H/C-X Coupling: Construction of Polycyclic Aromatic Hydrocarbons from Brominated Benzimidazoles.

A palladium/norbornene (NBE)-catalyzed 2-fold C-H/C-X coupling reaction of aryl iodides is reported. Bromine-substituted benzimidazoles were chosen as electrophilic and termination reagents, and the versatile polycyclic aromatic hydrocarbon products were successfully obtained. The strategy overcomes the challenge of catalyst poisoning by heterocyclic substrates.

Purpose: After successful endovascular treatment of acute ischemic stroke, there remains considerable controversy surrounding the efficacy of intensified blood pressure control therapy. Presently, numerous randomized controlled trials have yielded diverse findings. Thus, our objective is to consolidate all current randomized controlled trial data to evaluate whether intensified systolic blood pressure targets, in comparison to standard targets, offer superior safety and efficacy.

5-Aminopyrazole Dimerization: Cu-Promoted Switchable Synthesis of Pyrazole-Fused Pyridazines and Pyrazines via Direct Coupling of C-H/N-H, C-H/C-H, and N-H/N-H Bonds.

A Cu-promoted highly chemoselective dimerization of 5-aminopyrazoles to produce pyrazole-fused pyridazines and pyrazines is reported. The protocol generates switchable products via the direct coupling of C-H/N-H, C-H/C-H and N-H/N-H bonds, with the merits of broad substrate scope and high functional group compatibility. Gram-scale experiments demonstrated the potential applications of this reaction.

Complex problem solving (CPS) refers to a set of higher-order capacities that allow an individual to interact with a dynamic environment and solve complex problems. The purpose of this study was to develop and validate Sokoban, a game-based assessment of the planning-execution stage of the CPS framework proposed by PISA 2012. The psychometric properties of this instrument were examined in a large sample of Chinese students ( = 1145) ranging from elementary to tertiary education.

Supplemental oxygen therapy has been shown to reduce hypoxemia and is well tolerated in patients with ILD. However, little is known about the effect of nocturnal oxygen supplementation (NOS) on OSA in patients with ILD.

Dissection of the physiological interactomes of histone post-translational modifications (hPTMs) is crucial for understanding epigenetic regulatory pathways. Peptide- or protein-based histone photoaffinity tools expanded the ability to probe the epigenetic interactome, but in situ profiling in native cells remains challenging. Here, we develop a nucleus-targeting histone-tail-based photoaffinity probe capable of profiling the hPTM-mediated interactomes in native cells, by integrating cell-permeable and nuclear localization peptide modules into an hPTM peptide equipped with a photoreactive moiety.

Bisindole alkaloids constitute a significant class of natural compounds distinguished by their characteristic bisindole structure and renowned for their anticancer properties. Over the last six decades, researchers have isolated 425 microorganism-derived bisindole alkaloids (MDBAs). Among them, 187 MDBAs have demonstrated anticancer properties against various in vitro cancer cell lines, primarily by impeding the cell cycle, restraining cell proliferation, and inducing apoptosis and autophagy.

Asp-based lactam cyclic peptides are considered promising drug candidates. However, using Fmoc solid-phase peptide synthesis (Fmoc-SPPS) for these peptides also causes aspartimide formation, resulting in low yields or even failure to obtain the target peptides. Here, we developed a diaminodiacid containing an amide bond as a β-carboxyl-protecting group for Asp to avoid aspartimide formation.
