Background: Transgender and gender diverse (TGD) adults experience disability at twice the rate of cisgender (non-TGD) adults in the US. TGD people of color and low-income TGD people experience intersecting discrimination that may compound chronic conditions and disability. To our knowledge, no research has focused on chronic conditions among TGD Medicare beneficiaries with disabilities.

Acute myeloid leukaemia (AML) arising from a myeloproliferative neoplasm (MPN) is more aggressive and less responsive to therapies compared to de novo AML. Olutasidenib, an oral small-molecule inhibitor of mutated IDH1 (mIDH1), showed encouraging and durable responses in a phase 1/2 study of adults with post-MPN mIDH1 AML. Patients received olutasidenib 150 mg BID monotherapy or in combination with azacitidine.

Escape rooms are a gamification tool that are increasingly implemented in educational contexts. The aim of this scoping review was to examine the breadth and depth of how educational escape rooms are being used in health professions education, what educational goals are being achieved, and how these objectives are being assessed. Following PRISMA guidelines, a search of six databases using the key terms "escape room" and "health professional" was conducted.

Article Synopsis
  • Myelodysplastic syndromes (MDSs) treatment aims to improve patients' quality of life (QOL), especially concerning issues such as anemia and transfusion dependence.
  • A study involving 1120 MDS patients revealed that those who maintained transfusion independence (TI) had better overall survival (OS) and QOL compared to those remaining transfusion dependent (TD), while those switching between these statuses had intermediate results.
  • Specifically, patients who transitioned from TD to TI experienced improved QOL, while those moving to TD reported declines in their global QOL, especially in areas like fatigue and daily functioning.

A systematic literature review was conducted to determine the global status of newborn screening (NBS) for mucopolysaccharidosis (MPS) II (Hunter syndrome; OMIM 309900). Electronic databases were searched in July 2023 for articles referencing NBS for lysosomal storage diseases: 53 featured MPS II. Until recently, only Taiwan and two US states (Illinois and Missouri) formally screened newborns for MPS II, although pilot programs have been conducted elsewhere (Japan, New York, and Washington).

Article Synopsis
  • * This research identifies one tumor suppressor gene (NAGK) and five oncogenes (FXYD3, ACTR1A, B3GNT3, SIGIRR, and EXOC1) as significant in the early stages of pancreatic cancer using bioinformatic analysis and various databases.
  • * The study also finds that higher expression of the FXYD3 gene correlates with increased migratory ability in pancreatic cancer cell lines, suggesting its potential role in tumor progression and metastasis.

The oral route of drug administration is often preferred by patients and healthcare providers due to its convenience, ease of use, non-invasiveness, and patient acceptance. However, traditional oral dosage forms have several limitations, including low bioavailability, limited drug loading capacity, and stability and storage issues, particularly with solutions and suspensions. Over the years, researchers have dedicated considerable effort to developing novel oral drug delivery systems to overcome these limitations.

Adult acute myeloid leukemia (AML) patients under the age of 60 often receive similar intensive treatments, while outcomes between the adolescent and young adult (AYA) age group (18-39) and middle-aged adults (40-60 years) were seldom reported. We aim to study the characteristics and outcomes of AYA patients in comparison to middle-aged adults. A retrospective analysis was performed on AYA patients treated at Princess Margaret Cancer Center between 2008 and 2018.

Previous research has shown that morally-relevant theory of mind enables children to avoid blaming a peer for an accidental transgression. The current study investigated whether this form of theory of mind helps children recognize that gender inequalities are unfair and create negative emotional experiences. Further, the study examined this ability across three perspectives (for themselves, for those who have been advantaged by inequality, and for those who have been disadvantaged by inequality).

Introduction: Malignant hematology (MH) patients are susceptible to invasive fungal infections due to prolonged neutropenia and immunosuppressive therapies, which may require voriconazole therapy. Although voriconazole therapeutic drug monitoring (TDM) is common, evidence describing this practice is limited. The primary objective of this study was to describe the current practice of voriconazole TDM in MH patients at the Princess Margaret Cancer Centre (PM).

Mucopolysaccharidosis II (MPS II; Hunter syndrome; OMIM 309900) is a rare, X-linked, heterogeneous lysosomal storage disease. Approximately two-thirds of patients develop cognitive impairment, which is difficult to assess in clinical trials, partly owing to the variable nature of cognitive impairment. Analyzing data from siblings can help to minimize this heterogeneity.

Acute leukemia is a rapidly progressive cancer of the blood and bone marrow that requires a high degree of complex, specialized, resource-intensive clinical and supportive care. The aging Canadian population has introduced an unprecedented demand on the health care system for a variety of illnesses, including acute leukemia. The purpose of this work was to develop organizational requirements for service providers delivering care for patients aged 18 years and older with acute leukemia within a single-payer health care system in Ontario.

Article Synopsis
  • - Chronic hyperglycaemia in type 2 diabetes (T2DM) leads to vascular damage, and glycaemic variability (GV) plays a significant role in worsening health outcomes, though continuous glucose monitoring (CGM) data isn't widely used yet in T2DM management.
  • - A literature review was conducted to find high-level evidence linking GV metrics to vascular and clinical complications in T2DM, while excluding lower-quality studies that weren't as reliable.
  • - Six studies showed some association between GV indices (like standard deviation and time in range) and clinical issues, but results should be taken cautiously due to the moderate to low quality of the studies; more extensive and diverse research is needed to confirm these findings.

Article Synopsis
  • - The study investigates how NPM1 mutations affect the prognosis of acute myeloid leukemia (AML) that develops from prior chronic myeloid malignancies compared to de novo (new) AML cases.
  • - Out of 575 NPM1-mutated patients, only 51 (8.9%) had secondary AML (sAML), with an average time of 3.6 months from the initial chronic myeloid diagnosis to the development of sAML.
  • - Results showed no significant differences in leukemia-free or overall survival rates between de novo AML and sAML patients, suggesting that having a history of a chronic myeloid malignancy does not worsen prognosis for those with an NPM1 mutation.

Objective: Over 90 clinical trials demonstrate the efficacy of the collaborative care model (CoCM) to treat depression in primary care but there is significant variability in real-world CoCM implementation and scalability. This study aimed to determine the feasibility and effectiveness of an adapted CoCM in a safety-net primary care setting.

Methods: Bring It Up! (BIU) is a pilot trial comparing an adapted CoCM (intervention group) to usual care (historical controls) for primary care safety-net clinic patients with depression.

Introduction: Nonclinical evaluation of the cardiovascular effects of novel chemical or biological entities (NCE, NBEs) is crucial for supporting first-in-human clinical trials. One important aspect of these evaluations is the assessment of potential QT/QTc prolongation risk, as drug-induced QT prolongation can have catastrophic effects. The recent publication of E14/S7B Q&As allows for the situational incorporation of nonclinical QTc data as part of an integrated risk assessment for a Thorough QT (TQT) waiver application provided certain best practice criteria are met.
